摘要
目的 探讨体内、体外产生的小鼠桑葚胚玻璃化冻存后的发育能力。方法体内、体外产生的小鼠桑葚胚分别被含有乙烯甘醇、Ficoll和蔗糖的玻璃化溶液处理后冻存。结果体内产生的小鼠桑葚胚单纯暴露于玻璃化溶液中 ,而未冻存时其发育到胚泡期的百分率为 97 3% (一步法 )和 98 4% (二步法 ) ,与对照组 (98 1% )均无显著差异 (P >0 0 5 ) ,体外产生的小鼠桑葚胚单纯被玻璃化溶液处理后发育到胚泡期的百分率分别为 81 8% (一步法 )、82 4% (二步法 )和 83 6 % (对照组 ) ,三者之间无显著差异 (P >0 0 5 )。但是 ,体外产生的小鼠桑葚胚经玻璃化冻存融解后 ,发育到胚泡期的百分率为 70 6 % (一步法 )、81 3%(二步法 )和 83 6 % (对照组 ) ,一步法玻璃化组明显低于二步法玻璃化组和对照组 (P <0 0 5 ) ,而在体内产生的小鼠桑葚胚玻璃化冻存融解后 ,发育到胚泡期的百分率为 93 1% (一步法 )和 95 7% (二步法 ) ,二者之间无显著差异 (P >0 0 5 )。所有玻璃化冻存的小鼠桑葚胚融解后 ,显微镜下观察均未见透明带破裂。结论体内、体外产生小鼠桑葚胚胎经玻璃化冻存融解后 ,可保持较高的存活率和发育能力 ,二步法玻璃化更适合冻存体外产生的小鼠桑葚胚。
Objective To assess the developing potential of mouse morulae produced in vitro or in vivo after vitrification and evaluate the validity of one-step and two-step vitrification methods Methods Mouse morulae produced in vitro and in vivo were vitrified by the solution containing ethylene glycol, Ficoll and sucrose (EFS solution) with one-step and two-step methods The developing potential and status of zona pellucida in vitrified mouse morulae were assessed Results The percentages of morulae developed into blastocyst stage were 81 8% and 82 4%, or 97 3% and 98 4%, respectively, after one-step and two-step exposure of in vitro morulae or in vivo morulae to EFS solution alone, which did not show significant difference compared with their controls (P>0 05) The percentage of morulae developed to blastocyst in in-vitro morulae vitrified by one-step method significantly decreased compared with two-step method or control (70 6% vs 81 3%; 70 6% vs 83 6%, respectively, P<0 05) However, there was no significant difference between blastocyst rates of in vivo morulae vitrified by one-step and two-step methods (93 1% vs 95 7%, P>0 05) Fracture of zona pellucida was not observed in all thawed morulae after one-step and two-step vitrification methods, irrespective of in vitro morulae or in vivo morulae Conclusion s Morulae produced in vitro and in vivo after vitrification may maintain high survival rate and developing potential Two-step vitrification method with EFS solution was suitable for in vitro morulae, which improves the developing potential of in vitro morulae One-step and two-step vitrification methods had no detrimental effect on the developing potential of in vivo morulae
