二脱甲氧基姜黄素对体外HepG2 Caspase-3活性的影响 被引量:4

摘要 目的:研究二脱甲氧基姜黄素对体外培养人肝癌细胞HepG2 Caspase-3蛋白酶活性的作用。方法:MTT法检测二脱甲氧基姜黄素对体外培养人肝癌细胞HepG2的抑制作用;分光光度检测其对Caspase-3酶活性的影响;免疫组化法检测其对人肝癌细胞HepG2 Caspase-3蛋白的表达。结果:二脱甲氧基姜黄素对人肝癌细胞HepG2有明显的抑制作用(P<0.001)且呈剂量关系,与姜黄素有显著差异(P<0.001);二脱甲氧基姜黄素能增强Caspase-3酶活性,增强Caspase-3蛋白的表达。结论:二脱甲氧基姜黄素能激活Caspase-3蛋白活性,这可能也是它体外抑制肝癌细胞HepG2生长的机理之一。 Ojective : To study the effects of bisdemethyoxycurcumin on the activity of Caspase - 3 in HepG2. Method: MTI" was used to observe its effects on anti - proliferation of HepG2. The activity of Caspase - 3 was detected by spectro- photography. The Caspase - 3 protein expression was tested by immunohistochemical method. Result : The bisdemethyoxy- curcumin could inhibit the growth of HepG2 ceils (P 〈0.001 ) and it was more effective on anti - tumor than curcumin (P 〈 0. 001 ). It had significant effect on increasing Caspase- 3 expression. Conclusion: The bisdemethyoxycurcumin could inhibit the growth of HepG2. It can increase the expression of protein Caspase - 3. It can increase the activity of Caspase - 3, which may be one of its mechanisms associated with the inhibition of HepG2 cells growth.
