摘要
目的观察督脉、夹脊电针对脊髓损伤(SCI)后大鼠N-甲基-D-天冬氨酸受体亚基NR2b表达的影响。方法 SD大鼠32只随机分为假手术组、模型对照组、督脉电针组、夹脊电针组,用改良的Allen氏造模法建立大鼠SCI模型,两电针组取相应穴位于SCI后第8日开始每隔1 d进行1次电针治疗,共治疗6周。造模后1 d、1周、3周、7周对各大鼠后肢活动情况行BBB评分,术后7周取损伤区脊髓组织,Real-time PCR检测NR2b mRNA表达,Western-Blot检测NR2b蛋白的表达。结果大鼠下肢运动功能经BBB评分显示,两电针组评分与模组相比有明显升高(P<0.05);SCI后7周模型组NR2b表达较假手术组增高(P<0.05),治疗后两电针组NR2b表达较模型组下降(P<0.05),组间比较水平相近(P>0.05)。结论 SCI后电针可能通过降低脊髓组织NR2b表达量,减轻损伤的发生,改善下肢运动功能。督脉取穴与夹脊取穴治疗效果相近。
Objective:To observe the expression of NR2b subunit of N-methyl-D-aspartate receptor of SCI rats treated with electrical acupuncture on Dumai acupoint and Jiaji acupoint.Methods:32 SD rats were randomly divided into sham-operated group,model control group,electrical acupuncture on Dumai acupoint group and electrical acupuncture on Jiaji acupoint group,with 8 rats in each group.Modified Allen's method was used to establish spinal injury model.From the 8th day after SCI,the two electrical acupuncture groups were given treatment every other day,lasting 6 weeks.1 day,1 week,3 weeks,and 7 weeks after modeling,function of rats lower limbs were evaluated by Basso,Beatlie,Bresnahan (BBB) grading.7 weeks after operation,injured spinal tissue was obtained for NR2b expression detection by using Real-time PCR and Western-Blot.Results:BBB grading showed that scores increased significantly in electrical acupuncture groups than those in model control group (P< 0.05).7 weeks after SCI,expression of NR2b in sham-operated group was increased (P< 0.05).Compared with model control group,expression of NR2b reduced more obviously in electrical acupuncture groups (P< 0.05).No significant difference was detected between electrical acupuncture on Dumai acupoint group and electrical acupuncture on Jiaji acupoint group (P> 0.05).Conclusion:Electrical acupuncture treatment after SCI may improve motor function of the limbs through decreasing NR2b expression in injured spinal tissue.The choice of acupoints makes no difference.
