摘要
目的:探讨痰热清注射液(TRQ)对小鼠内毒素(LPS)性急性肺损伤(ALI)的保护作用及其机制。方法:30只健康清洁的ICR小鼠按随机数字表法分为生理盐水(NS)组、ALI组、TRQ组,每组10只。制备小鼠急性肺损伤模型;NS组气管内滴注生理盐水(2 ml·kg-1),模拟急性肺损伤过程;TRQ组于造模后立即给予TRQ4 ml·kg-1尾静脉注射,其余2组接受同剂量的生理盐水。各组均选择造模完成后6 h为观测点,观察每组肺指数、血清NO含量及动脉血气分析的结果;观察肺组织病理形态学改变;免疫组织化学检测肺Caspase-3的蛋白表达情况。结果:ALI组与NS组比较,肺指数升高(P<0.05);血清NO含量明显升高(P<0.01);PaO2显著下降(P<0.01);肺组织损伤明显,Caspase-3表达阳性率显著升高(P<0.01)。与ALI组比较,TRQ组肺指数降低(P<0.05);血清NO含量显著降低(P<0.01);PaO2明显升高(P<0.05);肺组织损伤程度较ALI组明显减轻,Caspase-3表达阳性率显著降低(P<0.01)。结论:NO在LPS诱导的ALI发病过程中起着重要作用,TRQ对LPS诱导的ALI的保护作用可能是通过降低血清NO含量,下调Caspase-3蛋白的表达有关。
OBJECrlVE To explore the protective effect and mechanism of Tanreqing injection (TRQ) on endotoxin (I.PS)- induced acute lung injury in mice. METHODS A total of 30 healthy and clean ICR mice were randomly divided into normal sa line groups (n= 10), ALI (n= 10) and TRQ-treated ALI groups (n = 10). A mice model of normal atmospheric temperature was established in the acute lung injury and TRQ-treated ALI groups, while normal saline group was simulated without use of LPS and intratracheal instillation with saline(2 ml~ kg-I) instead of; the mice in TRQ-treated ALI group were administered with tail vein injection of Tanreqing (4 ml. kg-1 ) immediately after the model was established, while the other two groups re- ceived the same volume of normal saline (0. 9%). Arterial and lung tissue samples were collected from 10 mice in each group at 6 h after the model was established. The content of serum NO and arterial blood gas levels were measured; lung tissue apopto- sis was evaluated, and expression of myocardial Caspase-3 was measured. RESULTS Compared with the NS group, in ALI group the lung index increased (P〈0. 05) ; the serum NO levels increased significantly (P〈0. 01 ) ; PaO2 declined significantly (P〈0. 01 ). Lung tissue damage was apparent,Caspase-3 expression positive rate was significantly increased (P〈0. 01 ). Compared with ALI group, the TRQ group decreased lung index (P〈0. 05) ; the serum NO levels decreased significantly (P〈 0. 01 );PaO2 increased significantly (P〈0. 05). The degree of lung tissue injury in the TRQ group was significantly reduced than that in ALl group, and Caspase-3 expression positive rate was significantly lower (P〈0. 01). CONCLUSION NO in the pathogenesis of ALI induced by LPS plays an important role in the process,TRQ exerts a protective effect on the lung tissue of mice with acute lung injury by lowering serum NO content, and reducing the Caspase-3 protein expression.
