摘要
为研究人参水提物(WEG)对SW480细胞体外增殖的抑制作用,采用倒置显微镜、MTT法观测SW480细胞的存活情况,并通过原子力显微镜获取各试验组细胞在人参水提物作用下细胞膜的平面/三维超微结构。结果表明:人参水提物能显著抑制SW480细胞增殖,其凋亡率与作用时间、剂量存在依赖关系(P<0.05),随着作用时间延长或剂量加大,对SW480细胞抑制作用呈增加趋势。原子力显微图像表明,正常SW480细胞呈圆形、细胞饱满,表面较光滑,而WEG处理组存活细胞皱缩,细胞表面的粗糙度较大,细胞膜存在一定的损伤。这说明WEG能显著抑制SW480细胞体外增殖,可能与WEG对SW480细胞膜的损伤有关。
The mechanism of inhibition and apoptosis of WEG on the vitro colorectal cancer SW480 cell lines was studied .The effects of different doses of WEG on the proliferation of the SW480 cell lines were investigated by inverted microscope,MTT assay and atomic force microscope (AFM).The images from these three observation methods show that WEG can reduce SW480 cell viability in a dose-dependent and time-dependent manner (P&lt;0.05).Those cells in the experimental groups were observed getting shrunk-en and round,membrane broken and surface rough,depending on the increasing dose .While the cells in the control group were round and full,the surfaces of the cells were smooth .The results show that WEG can inhibit the proliferation of SW480 cells significantly and this phenomenon may be caused by the injury of the cell membrane .
