摘要
【目的】建立玉叶金花药材高效液相色谱(HPLC)指纹图谱的分析方法。【方法】采用HPLC法,色谱柱为Diamonsil C18(2)(150 mm×4.6 mm,5μm),流动相条件为甲醇—体积分数0.5%冰醋酸溶液,梯度洗脱,流速:1.0 mL/min;检测波长265 nm;柱温25℃;记录时间65 min;使用中药色谱指纹图谱相似度评价系统(2004年A版)进行数据处理,对不同产地的玉叶金花药材指纹图谱的相似度进行比较分析。【结果】通过分析10批药材的HPLC图谱,确定了11个峰为指纹图谱的共有峰,构成了玉叶金花药材的色谱指纹图谱特征。【结论】所建立的方法精密度、稳定性和重复性良好,为该药材的鉴别及质量评价提供了新的依据。
Objective To establish an analysis method for fingerprints of Mussaenda pubescens Ait. f. by high performance liquid chromatography (HPLC) . Methods HPLC method was performed on a Diamonsil C ls (2) column (150 mm× 4.6 mm, 5 μm) with methanol-0.5% glacial acetic acid solution as mobile phase by gradient elution. The flow rate was 1.0 mL/min, the detection wavelength was 265 nm, column temperature was 25℃, and fingerprinting time lasted 65 minutes. The software of ”SOP of similarity evaluation system for chromatographic fingerprint of TCM (Version 2004A)“ was used to analyze the experimental data, and then the similarities of HPLC fingerprints of Mussaenda Pubescens from different habitats were compared. Results After analysis of the HPLC fingerprints of 10 batches of Mussaenda pubescens medicinal material, 11 peaks had been confirmed as common peaks, which constituted the chromatographic fingerprint characteristics of Mussaenda pubescens medicinal material. Conclusion The established method in this study is of higher precision, stability and reproducibility, which will provide evidence for the differentiation and quality evaluation of Mussaenda pubescens Ait. f..
