摘要
目的:观察三叶青乙酸乙酯提取物对裸鼠人肝癌细胞HepG-2皮下移植瘤的抑制作用,并探讨其相关作用机制。方法:采用腋下接种HepG-2细胞制备小鼠荷瘤模型,观察三叶青乙酸乙酯提取物低、中、高剂量对裸鼠肿瘤体积、瘤重、抑瘤率、脾指数的影响,生化分析仪检测生化指标ALT、AST、BUN、CRE,ELISA法检测血清中细胞因子TNF-α、IFN-γ的含量。结果:三叶青低、中、高剂量组的抑瘤率分别是38.66%、23.53%、31.09%;与模型组相比,三叶青各剂量组TNF-α、IFN-γ水平升高,其中高剂量组IFN-γ明显高于模型组,具有统计学差异。结论:三叶青乙酸乙酯提取物具有抑制HepG-2皮下移植瘤的作用,作用机制可能与提高IFN-γ和TNF-α水平相关。
Objective:To explore the antitumor effect and the mechanism of ethylacetate extract of tetrastigmatis hemsleyani in athymic mouse transplantation tumor by human hepatocarcinoma cell HepG-2.Methods:HepG-2 cell cultured in vitro was transplanted into subaxillas of mice to establish animal models with transplanted tumor.The tumor weight,volume,rates of tumor inhibition,spleen index were recorded,Diochemical analyzer analyzed ALT,AST,BUN,CRE,ELISA analyzed the levels of serum TNF-α and IFN-γ.Results:Low,moderate and high dose treatment groups showed the rates of tumor inhibition were 38.66%,23.53% and 31.09%.After treating with tetrastigmatis hemsleyani,all dose groups can up-regulated the levels of TNF-α and IFN-γ,and high dose treatment groups was significant difference to compared with model group.Conclusion:Ethylacetate extract of tetrastigmatis hemsleyani can inhibited athymic mouse transplantation tumor by human hepatocarcinoma cell HepG-2,its mechanism may be associated with the levels of TNF-α and IFN-γ.
