摘要
目的检测5-氟尿嘧啶(5-FU)对人胃癌细胞株MKN45及SGC-7901 p53,p53β表达的影响。方法将不同浓度5-FU作用细胞48h,用western-blot检测p53及P53β在蛋白水平表达的变化。结果 Westernblot结果显示,化疗药物5-FU作用MKN45胃癌细胞48h后,p53,P53β较未加药组表达增加,并且随药物浓度的增加其表达也随之增高,人正常胃黏膜细胞中p53,p53β的表达低于药物干预后的胃癌细胞;而相同条件下,在SGC-7901细胞系中,随药物浓度的增加p53,p53β的表达无明显变化。结论 5-FU能够抑制表达野生型p53人胃癌细胞系MKN45的增殖,并能上调p53,p53β的表达,推测p53β异构体参与的p53途径是5-FU诱导的肿瘤生长抑制的重要途径之一。5-FU能够抑制人胃中分化腺癌细胞SGC-7901的增殖,但与p53,p53β无明显关联。
Objective To explore the expression of p53β and its biological significance in 5-Fu treated gas- tric cancer cell lines. Methods MKN45 and SGC-7901 cells were cultured with 5-FU for two days,compare them with normal gastric mucosa cells and gastric; cancer cells without 5-FU ,western blot was used to detect the expression changes of p53,p53β. SPSS 17.0 software package was used for statistical analysis. P 〈 0.05 was determined to be significant difference. Results Western blot results showed that,with the increase concentration of 5-FU,the relative expression of p53 and p53β in MKN45 cells were increased remarkably,and the differences between the groups were statistically sig- nificant(P 〈0.05). In contrast,in SGC-7901 cell line,the expression of p53 and p53β were not change much with the increase of drug concentration. Conclusion In 5-FU can induce MKN45 gastric cancer cells proliferation inhibition,and the p53β plays as a cooperater with wide-type p53 ,inferring that p53β playes an important role in the inhibition effect of gastric cancer cell growth. The p53-p53β pathway may induce the proliferation inhibition in SGC-7901 cells, but the mechanism called for further exploration.
