摘要
目的探讨健脾益肠散对溃疡性结肠炎(UC)大鼠Toll样受体4(TLR4)的影响。方法将60只大鼠随机分为正常组、模型组、健脾益肠散高、中、低剂量组和柳氮磺吡啶组各10只;除正常组外,其余均采用2、4、6-三硝基苯磺酸(TNBS)/乙醇法复制UC大鼠模型;给药21d后,观察各组大鼠给药前后的体质量变化,用酶联免疫吸附测定法、免疫组织化学法分别检测各组大鼠血清TLR4水平和结肠组织TLR4蛋白表达情况。结果模型组大鼠体质量增长明显低于正常组(P<0.01),健脾益肠散高、中剂量组能明显促进UC大鼠体质量增长(P<0.05)。模型组大鼠血清TLR4水平和结肠组织TLR4蛋白表达均明显高于正常组(P<0.01);健脾益肠散高剂量组TLR4水平和高、中剂量组TLR4蛋白表达均较模型组降低,差异有统计学意义(P<0.05或P<0.01);健脾益肠散高剂量组对TLR4的影响与柳氮磺吡啶组比较差异不显著(P>0.05),但明显优于低剂量组(P<0.05)。结论健脾益肠散对UC肠粘膜免疫的保护作用可能与降低TLR4水平及其蛋白表达有关。
Objective To explore the effect of Jianpiyichang Powder(JP) on Toll - like receptor 4 ( TLR4 ) in ulcerative colitis (UC) rats. Methods 60 rats were randomly divided into the normal group, model group, JP high, medium and low- dose group, sulfasalazine group of 10 each. Besides normal group, the other used 2,4,6 - trinitrobenzene sulfonie acid (TNBS) / eth- anol method copied UC rat model. Observed body weight changes before and after drug administration, the serum of TLR4 level and colon tissue TLR4 protein expression were respectively detected by enzyme -linked immunosorbent assay and mmunohistochemis- try in each group of rats after gavage 21d. Results Body weight growth in model rats was significantly lower than that in normal group (P 〈0.01 ). JP high, middle dose group can effectively promote the growth of body weight of UC rats (P 〈 0.05 ). The model rats serum of TLR4 level and colon tissue TLR4 protein expression was significantly higher than that of the normal group( P 〈 0.01 ) ;TLR4 level in JP high dose group and the TLR4 protein expression in JP high , middle dose group compared with model group decreased significantly( P 〈 0.05 or P 〈 O. 01 ) ;JP high - dose group, the difference of TLR4 with sulfasalazine group was not significant( P 〉 0.05 ) , but significantly better than the low- dose group (P 〈 0.05 ). Conclusion Protective effects of JP on intestinal mucosal immunity of UC may be related to the decrease of TLR4 level and its protein expression.
