摘要
目的研究天麻素对化疗药致大鼠肝损伤的预防作用。方法用长春新碱隔日腹腔注射建立大鼠肝损伤模型,共5次;实验组每日腹腔注射不同剂量的天麻素(30,60,120 mg/kg),共9天,对照组和模型组用生理盐水同步对照,观察各组大鼠的一般状况;并于末次给药的第2天眼球取血,同时迅速取出肝脏,称湿重,计算肝指数(肝脏湿重/体重);检测大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST);并观察肝组织病理切片。结果与对照组比较,模型组的肝脏指数、ALT、AST和AST/ALT均明显升高(P<0.05);大鼠的毛发凌乱、活动减少、饮食和精神状况较差;肝组织病理切片显示肝索排列不规则,细胞有脂肪变性及气球样变等,甚至出现细胞核脱失,细胞坏死;而与模型组比较,各剂量天麻素组的上述指标均有不同程度的改善(P<0.05,P<0.01),其中高剂量天麻素的改善最明显,呈剂量依赖性。结论天麻素可剂量依赖性预防长春新碱引起的肝损伤,减轻其肝脏毒性。
Objective To study the preventive effect of gastrodin on chemotherapy drugs induced liver injury in rats. Methods - Vincristine was administered by intra -peritoneal injection on alternate days,5 times totally, to establish liver injury model. In the meantime, the experimental groups rats were injected different doses of gastrodin(30,60,120mg/kg), 9 days totally. The rats of control group and model group were injected saline synchronously. To observe the general situation of rats; At 10th day, to take blood from eyeballs and take out liver quickly and weigh the wet weight of liver and count the liver index( liver wet weight/body weight), to detect serum alanine aminotransferase ( ALT), aspartate aminotransferase ( AST), to observe liver tissue pathological slices. Results Compared with control group, in the model group, the liver index, ALT and AST and AST/ALT increased signifi- cantly (P 〈 0.05 ). The general condition showed that hair was messy, activities were decreased, diet and mental health were poor. Liver tissue pathology slices showed that hepatic cord were irregular and had eel1 fatty degeneration and ballooning change, cell nucleus attrition, cell necrosis. Compared with model group, in three gastrodin groups, the indexes were improved differently (P 〈0. 05 ,P 〈0.01 ), and it was the most obvious in the high dosage gastrodin group, which showed dose dependent. Conclu- sion Gastrodin can reduce hepatotoxicity, has obvious preventive effect on vincristine induced liver injury in rats, and shows dose dependent.
