摘要
目的:研究蛹虫草多糖(CMPS)对小鼠移植型腹水型肝癌(H22)体内诱导肿瘤细胞凋亡作用,探讨其抑瘤作用机制。方法:建立H22移植瘤小鼠模型,采用电镜技术观察H22荷瘤小鼠肿瘤组织的超微结构改变;免疫组织化学法检测蛹虫草多糖作用后H22肿瘤组织细胞内P53蛋白的表达情况。结果:电镜观察到蛹虫草多糖作用后H22肿瘤组织细胞有早期凋亡改变,肿瘤组织内有凋亡细胞存在;免疫组化实验结果显示,蛹虫草多糖作用后H22小鼠肿瘤组织细胞突变型P53蛋白的表达水平降低。结论:蛹虫草多糖具有诱导小鼠H22肝癌细胞凋亡作用,促凋亡作用可能与其降低H22小鼠肿瘤组织细胞内突变型P53蛋白的表达有关。
Objective :To evaluate the effects of Cordyceps militaris polysaccharides(CMPS) on apoptosis of tumor transplanted with hepatoma cells in mice(H22) in vivo and explore the anti-tumor mechanism. Methods:Establish H22 xenograft mouse model,electron microscope was used mehod to observe the ultrastructural changes of H22 in tumor bearing mice. P53 protein expression in H22 tumor cells was detected by immunohistochemistry method. Results:CMPS could reduce apoptosis in H22 hepatoma cells in mice. Conclusion:The apoptosis may be related to its decreasing mutant P53 protein expression in tumor tissue in mice.
