响应面法优化头孢菌素C摇瓶发酵培养基 被引量:3

Improving the fermentation yield of cephalosporin C by response surface analysis

摘要 目的优化头孢菌素C发酵培养基,提高发酵水平。方法采用Plackett-Burmana法、最陡爬坡试验和中心组合设计,用摇瓶发酵进行试验,利用minitab15.0软件进行二次回归分析。结果 Plackett-Burman设计证明蔗糖(A)、玉米浆(B)和硫酸铵(C)对效价影响较大,通过最陡爬坡试验和响应面法得到最佳发酵培养基配方为:蔗糖1.49%,玉米浆17.98%,硫酸铵0.83%。结论使用响应面法可以显著提高头孢菌素C的发酵效价。 Objective To optimize the culture medium and improve the yield of cephalosporin C fermentation. Methods Plackett-Burman design is used to select the most influencing factors determination, the path of steepest ascent for the center test point determination, the central composite designed for the response surface analysis, experimented by shaking flask for fermentation. Minitab 15.0 software for analysis. Results The three factors that selected by Plackett-Burman method are sucrose(A), corn steep liquor(B) and ammoniuin sulfate(C) which are the most important factors for the production of cephalosporin C. The optimum medium for cephalosporin C production is sucrose 1.49%, corn steep liquor 17.98%, ammonium sulfate 0.83%. Conclusion Response surface analysis is a good combination for improving cephalosporin C fermentation yield.
