摘要
研究建立了一种采用水蒸气蒸馏法提取各种食品中的脱氲乙酸,然后用高效液相色谱仪进行检测的方法。实验采用C18色谱柱(5μm,250mm×4.6mm),流动相为甲醇+0.02mol/L乙酸铵(体积比为5+95),柱温30℃,检测波长为30611112。标准曲线有良好的线性关系(r=0.9996),方法检出限为0.008g/kg,加标回收率为92.6%~104%,对不同的样品基质相对标准偏差为0.69%-3.53%。该方法样品处理步骤简单,无有机溶剂消耗,方法的回收率、灵敏度和准确度高,精密度良好,重现性好,尤其适用于高脂、高油样品的前处理,更能满足样品的分析要求。
A method for the extraction of dehydroacetic acid in various foods by steam distillation method was established. And then dehydroacetic acid was determinated by high performance liquid chromatographic (HPLC). Separation was carried out on a column of C18 (5 μm,250mm×4.6mm) at the temperature of 30 ℃ with methanol +0.02 mol/L ammonium acetate (v/v, 5+95) as mobile phase. The detection wavelength was 306 nm. The results showed a good linear relationship between the peak area and the concention (r=0.9996), the recoveries for dehydroacetic acid products were 92.6%-104%, and the relative standard deviation was 0.69%-3.53% for different kinds of food. The limit of detection was approximately 0.008 g/kg. The procedure described here was simple, selective, accurate, without of organic solvent consumption. The pretreatment method was especially suitable for high fat and high oil samples to better meet the requirements of sample analysis.
