摘要
目的研究耐药肺结核患者外周血中CD+4CD+25CD127low调节性T细胞及IL-17的表达水平与非耐药肺结核患者、健康对照者的差异。方法检测40例耐药肺结核患者(耐药组)、40例非耐药肺结核患者(普通组)和40例健康对照者(健康对照组)外周血CD+4CD+25CD127low调节性T细胞表达强度;并采用酶联免疫吸附试验(ELISA)检测辅助性T细胞17(Th17)分泌的细胞因子IL-17表达水平。结果耐药组CD+4CD+25CD127low调节性T细胞表达为(7.85±2.03)%,普通组为(6.23±1.88)%,健康对照组为(5.14±1.67)%,三组之间差异显著(P均<0.01);耐药组IL-17含量为(14.29±6.71)ng/L,而普通组为(19.64±7.82)ng/L,健康对照组为(25.42±8.67)ng/L,三组之间差异显著(P均<0.01)。结论外周血CD+4CD+25CD127low调节性T细胞、IL-17水平在耐药肺结核患者和非耐药肺结核中表达的差异可能是耐药机制产生的原因之一,与耐药性的形成或严重程度有相关。
Objective To study the difference in CD4+ CD25+CD127 and IL-17 in peripheral blood among the drug-resistant tuberculosis patients,non-drug-resistant tuberculosis patients and healthy people.Methods Flow cytometry was applied to detect the expression percentage of peripheral blood CD4+ CD25+ CD127 in drug-resistant tuberculosis patients (the drug-resistant group,n =40),non-drug-resistant tuberculosis patients (the non-drug-resistant group,n =40) and healthy people (the healthy control group,n =40).Enzyme-linked immunosorbent assay (ELISA) was used to detect the level of IL-17 among the three groups.Results The percentage of CD4+CD25+ CD127/CD4+ in the drug-resistant group accounted for 7.8 ± 2.03%,6.23 ± 1.88% in the non-drug-resistant group,and (5.14 ± 1.67)% in the control group (P <0.01).The levels of IL-17 in the drug-resistant group accounted for (14.29 ± 6.71) ng/L,(19.64 ± 7.82) ng/L in the non-drug-resistant group,and (25.42 ± 8.67) ng/L in the control group (P < 0.01).Conclusion There are significant differences for the indexes percentage of CD4+ CD25+ CD127 and the level of IL-17 between drug-resistant and non-drug-resistant TB patients,which may be one of reasons for drug-resistant and involve in the formation and the severity of the drug-resistant in TB patients.
