摘要
目的:采用高效液相色谱法测定风湿止痛药酒(FSZTYJ)中粉防己碱、防己诺林碱、蛇床子素和二氢欧山芹醇当归酸酯的含量。方法:HypersilC18色谱柱(4.6Fmm×150mm,5μm);流速:1.0ml·min^-1;粉防己碱和防己诺林碱:流动相为甲醇-乙腈-0.3%二乙胺(52:37:11),检测波长:280nm。蛇床子素和二氢欧山芹醇当归酸酯:流动相A为甲醇,流动相B为0.2mol·L^-1硫酸铵溶液,检测波长:322nm。结果:粉防己碱和防己诺林碱分别在0.1026-2.052μg(r=0.9991)和0.0694~1.388ug(r=0.9995)时进样量与峰面积呈良好的线性关系,平均加样回收率分别为97.30%和96.93%,RSD(n=6)分别为1.44%和1.35%;蛇床子素和二氢欧山芹醇当归酸酯分别在0.0572~1.144μg(r=0.9992)和0.0196~0.392μg(r=0.9994)时进样量与峰面积呈良好的线性关系,平均加样回收率分别为98.19%和96.60%,RSD(n=6)分别为1.50%和1.53%。结论:该方法测定结果准确、灵敏、重复性好。
OBJECTIVE To develop an HPLC method for determination of the content of tetrandrine, fangchinoline, osthole and columbianadin in FSZTYJ. METHODS A Hypersil C18 column was used as the chromatographic column,the flow rate was 1.0 ml,min . Tetrandrine and fangchinoline..The mobile phase consisted of methanol-acetonitrile-0. 3% diethylamine(52:37: 11). The UV detection wavelength was at 280 nm. Osthole and columbianadin: The mobile phase A consisted of methanol, the mobile phase B consisted of 0. 2 mol · L^- 1 ammonium sulfate solution, The UV detection wavelength was set at 322 nrrL liE- SULTS There was a good linear relationship between the concentration of tetrandrine, fangchinoline and peak area value when the concentrations of tetrandrine and fangchinoline were within the range of 0. 102 6- 2. 052μg(r= 0. 999 1),0. 069 4- 1. 388 μg(r= 0. 999 5). The average recovery was 97. 30%(RSD= 1. 44%) and 96. 93 % (RSD= 1.35%). There was a good linear re- lationship between the concentration of osthole, columbianadin and peak area value when the concentrations of osthole and co- lumbianadin were within the range of 0. 057 2- 1. 144/lg(r= 0. 999 5),0. 019 6- 0. 392μg(r= 0. 999 4). The average recovery was 98. 19%(RSD= 1.50%) and 96. 60%(RSD= 1.53%). CONCLUSION The method was accurate,sensitive,good repeat- ability and could be used for the determination oftetrandrine, fangchinoline, osthole and columbianadin in FSZTYJ.
