摘要
目的 观察心肌缺血再灌注时心肌细胞凋亡现象 ,研究Bcl- 2、C -myc蛋白的表达情况。方法 采用末端标记技术 (TUNEL)标记凋亡细胞 ,免疫组织化学法检测Bcl- 2、C -myc蛋白的表达。结果 在持续缺血和缺血再灌注大鼠心肌中 ,TUNEL法检测到大量的阳性细胞。随着再灌注时间的延长 ,大鼠心肌细胞凋亡数逐渐增多。持续缺血 4 5h组的心肌细胞凋亡数明显高于缺血 30min再灌注 4h组的凋亡心肌细胞数。免疫组织化学法检测发现 ,与假手术组比较 ,持续缺血 4 5h组和缺血 30min再灌注 4h组Bcl- 2、C -myc蛋白的表达都增加 ,缺血再灌注组中Bcl- 2蛋白表达明显上调 ,C -myc蛋白表达明显下调。结论 心肌缺血再灌注中存在细胞凋亡现象 ,且心肌细胞凋亡与Bcl- 2、C -myc蛋白的表达密切相关。
Objective To study the significance of myocyte apoptosis in myocardial ischemic reperfusion (MIR) in rats.To evaluate the relationship between Bcl-2 and C-myc and myocyte apoptosis in MIR.Methods Myocyte apoptosis in MIR rat model was studied by TdT-mediated dUTP nick end labeling (TUNEL).Bcl-2 and C-myc was studied by immunohistochemical staining.Results Continuous ischemia as well as ischemia followed by reperfusion can result in myocyte apoptosis in rat myocardium.The permillage of apoptotic myocytes increased with the duration of reperfusion.We demonstrate that the fraction of cardiomyocytes undergoing apoptosis is lower in the reperfused heat than in the continuously ischemic heat.We demonstrate that the expression of Bcl-2 is higher in the reperfused heat than in the continuously ischemic heat and the expression of C-myc is lower in the reperfused heat than in the continuously ischemic heat.Statistical analysis demonstrated a negative correlation between the pemillage of apoptotic myocytes and Bcl-2,and a positive correlation between the permillage of apoptotic myocytes and C-myc.Conclusion The elucidation of cardiac apoptosis mechanisms will provide useful information that could potentially aid in the understanding and treatment of myocardial ischemic reperfusion injury.
