摘要
目的 观察糖皮质激素 (Dex)、孕激素 (P)、雌激素 (E)、雄激素 (T)等甾体激素 (10 -10 ~ 10 -6mol/L)对人卵巢癌细胞系 3AO增殖及分化的影响。方法 以不同浓度Dex、E、P、T处理培养的 3AO细胞 ,采用活细胞计数观察细胞增殖情况 ,用氨基氨替比林法测定碱性磷酸酶 (ALP)活性 ,用酶联免疫法测定 3AO细胞标志抗原CA12 5水平的变化 ;采用完整细胞饱和测定法测定 3AO细胞中糖皮质激素受体 (GR)的表达。结果 Dex对3AO细胞的增殖有明显的抑制作用 ,糖皮质激素受体拮抗剂Ru486可以阻断糖皮质激素对 3AO的增殖抑制作用 ;E、P、T等甾体激素对 3AO细胞的增殖无明显作用 (P >0 .0 5 )。Dex作用后ALP活性较对照明显增加 ,CA12 5水平明显降低 (P <0 .0 0 1) ;而E、P、T作用后ALP活性无明显变化 (P >0 .0 5 ) ,CA12 5较对照组明显下降(P <0 .0 0 1)。 3AO细胞含有可饱和性高亲和力GR ,最大结合容量为 92 .2 6± 2 3.30fmol/ 10 -6cells ,平衡解离常数为 1.37± 0 .19nmol/L ,特异结合实验表明 ,GR只与糖皮质激素特异结合。结论 糖皮质激素对 3AO细胞系有增殖抑制和诱导分化的作用 ,其调节作用通过GR来介导 ;E、P、T等甾体激素对 3AO细胞无增殖抑制作用。
Objective To observe the effects of glucocorticoids,estrogen, progesterone,androgen, testosterone on the proliferation and differentiation of 3AO cells. Methods 3AO cell proliferation was evaluated by viable cell count, alkaline phosphatase (ALP) activity was determined as described and expression was measured by ElISA. GR expression was studied with the standard radioligand binding assay. Results Dex could inhibit the proliferation of 3AO cells, but the cellular effects of glucocorticoids on 3AO cells could be reversed by RU486,a potent glucocorticoid antagonist. Sex steroid hormones had little effects on the proliferation of 3AO cells. ALP activity increased significantly after treatment with Dex and had no effects with sex steroid hormones. Furthermore, Dex and sex steroid hormones could suppress the expression of tumor maker CA125 in 3AO cells. Binding studies showed that there existed a saturable, high affinity GR in 3AO cells, with a maximum binding capacity of 92.26±23.30 fmol/10 6 cells, and a constant dissociation value of 1.37±0.19 nmol/L. Conclusion Dex can inhibit the proliferation and induce the differentiation of 3AO cells, and the cellular effects of glucocorticoids on 3AO cells is mediated by GR; but sex steroid hormones has no effects on the proliferation of 3AO cells.
