摘要
目的观察大鼠低氧脑损伤环境下补体C5a对大脑皮质细胞分泌IL-6的调节作用。方法选用成年体质量均等的雄性大鼠54只,分为1%低氧建模组(A组)18只、4%低氧建模组(B组)18只和常氧量对照组(C组)18只。各组在相应环境下处理3 h后,采用real-time PCR技术,检测建模大鼠给予抗C5a单克隆抗体以及外源性C5a后IL-6分泌水平。结果 A、B组较C组补体C5a水平明显升高(P均<0.05),IL-6水平降低。给予等量抗C5a单克隆抗体后,A组IL-6明显低于B、C组。给予外源性C5a后,A组IL-6水平明显高于B、C组。结论补体C5a在大鼠低氧脑损伤时对IL-6分泌有调节作用。
Objective It is to observe the regulating effect of complement C5a on the secretion of IL-6 by cerebral cortical cells in rats with hypoxic brain injury.Methods 54 adult male rats with equal weight were divided into 1% hypoxic model group,4% hypoxic model group and normal oxygen control group,each group had 18 rats.The level of IL-6 was detected by real-time PCR technology in the rats after modeling under corresponding conditions and giving anti-C5a monoclonal antibody and exogenous C5a.Results The levels of complement C5a were significantly higher and the level of IL-6 was lower in group A and group B than that in group C (P 〈 0.05).After giving anti-C5a monoclonal antibody,the level of IL-6 in group A was lower than that of group B and C.After giving exogenous C5a,the level of group A was higher than that in group B and C.Conclusion Complement C5a has regulation effect on IL-6 secretion in rats with hypoxic brain injury.
