摘要
目的:测定加味丹参饮中丹参酮ⅡA和丹酚酸B的含量。方法:采用HPLC法,色谱柱:(4.6 mm×250 mm,5μm);丹参酮ⅡA:流动相为甲醇-水(75∶25);流速:1 mL·min-1;柱温:30℃;λ=270 nm;丹酚酸B:流动相:甲醇乙腈-甲酸-水(30∶10∶1∶59);流速:1 mL·min-1;柱温:30℃;λ=286 nm。进样量:10μL。结果:丹参酮ⅡA和丹酚酸B分别在0.016-0.32μg,0.14-2.8μg内呈良好的线性关系;平均回收率分别为101.2%(RSD%=2.47%,n=5),99.6%(RSD%=2.71%,n=6)。结论:该方法准确、可靠、重复性好,可用于加味丹参饮的质量控制。
Objective: To determine the content of Tanshinone IIA and SalVianolic Acid B in the Jiawei Danshen Decoction. Methods: The contents of Tanshinone IIA and SalVianolic Acid B in the Jiawei Danshen Decoction are determined by HPLC method. Chromatographic column (4.6 mm×250mm,5μm), for Tanshinone IIA, mobile phase (Methanol-water 75:25); flow rate 1 mL·min-1; column temperature 30 ℃ ;h=270 nm; for SalVianolic Acid B, mobile phase (Methanol acetonitrileformic acid - water 30:10:1:59); flow rate 1 mL·min-1; column temperature 30℃;h=286 nm; sample size of 10μL. Results: The content of Tanshinone IIA and SalVianolic Acid B are respectively 0.016-0.32μg and 0.14-2.8 μg which showed well linear relation. The average recoveries (n=5) of Tanshinone IIA and SalVianolic Acid B were 101.2%(RSD%=2.47%,n=5) and 99.6% (RSD%=2.71%,n=6), respectively. Conclusion:The method is accurate, reliable and well iterancy. This method can use to control the quality of Jiawei Danshen Decoction.
