摘要
目的:研究芎麻汤对硝酸甘油诱导的偏头痛模型大鼠血浆及三叉神经颈髓复合体(TCC)上降钙素基因相关肽(CGRP)表达的影响,初步探讨芎麻汤的作用靶点。方法:36只雄性SD大鼠随机分为6组,分别为空白组,模型组,芎麻汤高、中、低剂量组,舒马曲普坦组,每组6只;采用皮下注射硝酸甘油(10mg/kg)复制偏头痛模型,给药后取颈外静脉血采用ELISA试剂盒检测血浆中CGRP含量,心脏灌注后取大鼠TCC部位采用免疫组织化学检测CGRP的表达。结果:与模型组比较,芎麻汤能有效降低偏头痛大鼠血浆中CGRP含量,并有效降低偏头痛大鼠TCC上CGRP免疫阳性染色光密度值,差异具有统计学意义(P<0.05,P<0.01)。结论:芎麻汤对硝酸甘油诱导的偏头痛模型大鼠具一定的治疗作用,其机制可能与调节CGRP水平有关。
Objective: To study the effects of Xiongmatang on CGRP expression of plasma and TCC on animal model of Migraine with Nitroglycerin, to discuss the targets of Xiongmatang. Methods: 36 SD rats were randomly divided into 6 groups: blank group, model group, high, middle and low dose of Xiongmatang groups, and Sumatriptan group. By giving hypodermic injection of 10mg/kg nitroglycerin to copy the migraine model. To observe the content of CGRP in plasma by ELISA method, and the expression of CGRP in the TCC by immunohistochemistry. Results: Xiongmatang could effectively decrease the content of CGRP in plasma and positive immunostaining OD of CGRP in TCC. Compared with the model group, the difference was significant (P〈0.05, P〈0.01). Conclusion: Xiongmatang was effective on the migraine rats induced by nitroglycerin, the mechanism may be related to adjust the level of CGRP.
