摘要
通过2,4-二氨基-6-(2′-吡啶基)均三嗪和高氯酸铜髤反应合成了一个配合物:[Cu(H2O)(PyTA)2](ClO4)2[PyTA=2,4-二氨基-6-(2′-吡啶基)均三嗪]。通过元素分析、红外光谱、紫外可见光谱、摩尔电导率和X-射线单晶衍射等方法对其进行了表征。结果表明该配合物晶体属单斜体,P21/c空间群,其晶体学参数:a=0.980 24(6)nm;b=1.248 31(7)nm,c=2.157 27(11)nm;β=108.657(3)°;Z=4,V=2.501 0(2)nm3,R1=0.054 3,wR2=0.1506。另外,应用试管二倍稀释法测定了配合物的抗菌活性,通过电子吸收光谱、荧光光谱、粘度测定及琼脂糖凝胶电泳等方法研究了配合物与DNA的作用。结果表明,与2,4-二氨基-6-(2′-吡啶基)均三嗪相比较,该配合物具有良好的抗菌活性,以插入模式与CT-DNA作用,并且在抗坏血酸存在下通过羟基自由基·OH切割pBR322DNA。
A copper(H) complex: [Cu (HEO)(PyTA)z](CI04)2 [PyTA=2,4-diamino-6-(2'-pyridyl)-l,3,5- triazine], was synthesized and characterized by elemental analysis, molar conductivity, IR, UV-Vis and single crystal X-ray diffraction. The crystal belongs to monoclinic, space group P21/c, with the crystal cell parameters: a=0.980 24(6) nm, b=1.248 31(7) nm, c=2.157 27(11) nm;/3=108.657(3)°; Z=4,V=2.501 0(2) nm3,Rl=0.054 3, wR2=0.150 6. The complex was assayed against gram-positive (S.aureus, B. subtilis, B. thuringiensis) and gram-negative (X. oryzae, Salmonella, E. coil)bacteria by doubling dilutions method, and the interaction of the complex to DNA was investigated by electronic absorption, fluorescence, viscosity measurement and agarose gel electrophoresis assay. The results showed that the complex has good antibacterial activities, and could bind to CT-DNA by an intercalative mode, and cleave pBR322 DNA by hydroxyl radicals as the active species in the presence of Vit C.
