摘要 将碱性蛋白酶用于提取废弃羊毛中的角蛋白。探讨了在羊毛角蛋白提取过程中,预处理双氧水用量、预处理温度和时间、pH值、蛋白酶用量及蛋白酶处理时间对羊毛溶解率的影响。优化的提取工艺为:预处理为H2O2 15 g/L,80 ℃,1 h,pH=10.5;酶解反应为碱性蛋白酶3%(omf),60 ℃,2 h,pH=9~10。该条件下羊毛的溶解率为88.9%,角蛋白产率为86.2%。对提取到的角蛋白进行红外光谱、X射线衍射和热重测试,结果表明,预处理中羊毛结构中部分二硫键断裂,角蛋白的结晶度显著增高,达到45.22%,热稳定性相对变差。 The alkaline protease is applied to the extraction of the waste wool keratin. The influences of the dosage of hydrogen peroxide, pretreatment time, temperature and pH value, dosage of protease and protease treatment time on the final dissolving rate of wool are discussed. The optimum extraction process is pretreatment at 80 ℃ for 1 h with H2O2 15 g/L, and enzymatic hydrolysis at 60 ℃ for 2 h with pH value of 9~10 and alkaline protease 3%(omf).The final dissolving rate of wool is 88.9% and the yield of keratin is 86.2% under above conditions. The results of infrared spectrum, X ray diffraction and thermo gravimetric of the extracted keratin show that part of disulfide bonds rupture during the pretreatment, the crystallinity of the keratin is higher up to 45.22%, and the thermal stability is relatively poor.
