新疆地区冠心病秽浊痰阻证动物模型的实验研究 被引量:2

摘要 目的通过人工气候箱结合喂养高脂饲料并腹腔注射维生素D3,建立新疆地区冠心病秽浊痰阻证大鼠模型。通过组织切片、血清指标,摸索模型建立条件。方法按RCT原则,将20只SD大鼠分为空白对照组、秽浊痰阻证模型组,每组10只。空白对照组正常饮水喂基础饲料;秽浊痰阻证模型组正常饮水喂高脂饲料并腹腔注射维生素D3,同时每日固定11 h在人工气候箱寒燥条件下饲养。实验第13周,行大鼠主动脉瓣至髂肾动脉分支处动脉及肝脏组织切片检查;测体质量、血清总胆固醇、三酰甘油、高密度脂蛋白胆固醇、低密度脂蛋胆固醇、极低密度脂蛋白胆固醇、血清白细胞介素6、肿瘤坏死因子-α。结果两组大鼠体质量、血清总胆固醇、低密度脂蛋白胆固醇、极低密度脂蛋白胆固醇、IL-6、TNF-α差异均有统计学意义(P<0.05),而三酰甘油、高密度脂蛋白胆固醇差异无统计学意义(P>0.05);动脉血管壁内膜下见无定形物质沉积,有泡沫样细胞;肝组织外周出现中度肝脂肪变性。结论冠心病秽浊痰阻证大鼠模型,基本表现出新疆地区冠心病秽浊痰阻证辨证要点,初步推测造模方法具有一定可行性。 Objective: To establish animal model of Xinjiang coronary heart disease with phlegm-turbidity syn- drome by combined treatment of artificial climate chamber,high-fat diet,abdominal injection of vitamin D3,and to explore animal model conditions by biopsy and serum index. Methods: By RCT principle, 20 SD rats were divided into normal control group and phlegm-turbidity syndrome group, 10 rats in each group. Normal control group was fed the basis diet;phlegm-turbidity syndrome group was fed the high-fat diet,injected with vitamin D3,and fixed 11 hours a day in the artificial climate chamber of cold dry conditions.13 weeks later,tissue sec- tions of aortic renal artery branch to iliac artery and liver were checked. Then weight, total cholesterol in serum, triglycerides,high-density lipoprotein cholesterol,low-density lipoprotein cholesterol,extremely low density lipoprotein cholesterol,interleukin-6in serum, and tumor necrosis factor-or were measured. Result: Compared with normal control group,body weight,total cholesterol in serum,low-density lipoprotein cholesterol,extremely low density lipoprotein cholesterol,serum interleukin-6,and tumor necrosis factor-eL in model group had statistical differences (P〈 0.05). In aspects of triglycerides and high-density lipoprotein cholesterol,there were no signifi- cant differences (P〉 0.05). There were amorphous materials deposited under artery wall intima. Foam-like cells and moderate hepatic steatosis in liver tissue periphery were observed. Conclusions: Animal model demonstrates the basic clinical characters of Xinjiang coronary heart disease with phlegm-turbidity syndrome so modeling method is initially speculated of certain feasibility.
