摘要 目的探讨Skp2和Cx43蛋白在正常宫颈上皮(NEC)、上皮内瘤样病变(CINI、CINⅡ、CINⅢ)及宫颈鳞癌(SCC)中的表达及其意义。方法采用免疫组化(S-P)法检测20例NCE、28例CINⅠ、24例CINⅡ、27例CINⅢ、58例SCC中Skp2和Cx43蛋白的表达情况,统计分析其表达的差异性及与宫颈癌临床病理因素的关系,探讨两者间的相关性。结果 Skp2在NCE、CINⅠ、CINⅡ、CINⅢ和SCC中阳性表达率逐渐增高(分别为10.00%、46.43%、45.83%、70.37%和75.86%),NCE组与其他各组比较,差异均有显著性(P<0.05);Cx43阳性表达率逐渐降低(分别为100%、100%、100%、74.07%和58.62%),NEC与SCC比较,差异亦有显著性(均P<0.05)。宫颈癌中Skp2表达与宫颈间质浸润深度、盆腔淋巴结转移有关;在不同宫颈组织中Skp2和Cx43表达呈负相关性(r=-0.894,P=0.041)。结论Skp2表达随宫颈病变加重而逐步升高,Cx43表达逐渐降低。Skp2表达与宫颈癌预后有一定关系。 Objective To explore the expressions and clinical significances of Skp2 and Cx43 in normal cervical epithelium,CINⅠ,CIN Ⅱ,CIN Ⅲ and cervical squamous cell.Methods The Skp2 and Cx43 expression in 20 cases of NCE,28 cases of CIN Ⅰ,24 cases of CIN Ⅱ,27 cases of CIN Ⅲ,58 cases of SCC tissue was analyzed by immunohistochemistry and the association of Skp2 and Cx43 expression with clinicopathological characteristics,tumour grade,deeper penetration,and metastasis of cervical carcinoma cells were evaluated.Results Skp2 expression was gradually increased in NCE,CINⅠ,CIN Ⅱ,CIN Ⅲ and SCC (10.00%,46.43%,45.83%,70.37%,75.86%,P<0.05),whereas Cx43 expression was gradually reduced with the increase in cervical lesions (100%,100%,100%,74.07% and 58.62% ; P< 0.05).Increased Skp2 staining was correlated with an increased aggressiveness of the tumor,including deeper penetration,and metastasis of cervical carcinoma cells.In contrast,the expression of Cx43 was negatively correlated with the expression of Skp2 (r =-0.894,P=0.041).Conclusion These findings suggest that Skp2 expression was gradually increased in cervical lesions,whereas Cx43 expression was gradually reduced.The Skp2 expression corresponds with higher stage tumours in cervical carcinoma,thus indicating a poor prognosis for patients.
