摘要
从土壤中筛选1株能产生右旋糖酐酶的菌株DN01,对该菌进行了分类鉴定和发酵条件考察。结合菌落形态、孢子形态特征和菌株的ITS rDNA序列分析进行菌株的鉴定,确定该菌株属于拟青霉(Paecilomyces)。在30℃120r/min摇床振荡培养8d后,发酵液上清中的右旋糖酐酶活力最高达到19.34U/ml。通过单因素试验对发酵条件进行考察,结果表明产生右旋糖酐酶的最适碳源为右旋糖酐,最适氮源为硫酸铵,培养基的起始pH为4.O~5.0,最适温度为30℃。
A dextranase-producting strain was screened.Strain DN01was identified and the fermentation of dextranase was studied.On the basis of morphological observation and ITS rDNA sequence analysis,strain DN01was identified as Paecilomyces.The yiele of dextranase activity was up to 19.34U/ml after growth at 30℃for 8days on the shaker with speed of 120r/min.Single factor experiment showed the optimal carbon source was dextran;the optimal nitrogen source was(NH4)2SO4;the optimal initial pH was4.0~5.0,and the optimal temperature was 30℃for dextranase producing.
