摘要
目的采用藻酸盐凝胶为支架进行肝癌HepG2细胞的体外培养,构建体外肝癌细胞三维培养模型。方法将HepG2细胞接种于藻酸盐凝胶中进行培养,通过倒置显微镜观察、H-E染色、钙黄绿素-碘化丙啶荧光复染及MTT实验,了解藻酸盐凝胶内细胞的生长情况。结果在藻酸盐凝胶中HepG2细胞以细胞团的形式生长,增殖活跃。结论成功建立了肝癌HepG2细胞藻酸盐凝胶培养体系,且该体系有利于HepG2细胞的生长。
Objective To establish the alginate three -dimensional culture system for human HepG2 cells. Methods HepG2 cells were encapsulated in algiuate hydrogel for three - dimensional culture. Cell morphology and growth in gel was demonstrated by microscopy and hematoxylin -eosin staining, Calcein -AM/PI staining, and MTF assay, respectively. Results HepG2 cells grew well when cultured in alginate gel. Conclusion The algi- nate three -dimensional culture system for human HepG2 cells was successfully established, and the microenvi- ronment provided by alginate hydrogel can contribute to the proliferation of HepG2 cells.
