瑞舒伐他汀改善大量饮酒对急性心肌梗死大鼠血管新生的影响 被引量:5

Rosuvastatin reverses the effect of heavy drinking on angiogenesis after infarction in rats

摘要 目的探讨大量饮酒对急性心肌梗死后血管新生的影响以及瑞舒伐他汀的干预作用。方法取雄性SD大鼠24只,随机分成3组:空白对照组、大量饮酒组[50%食用乙醇,5 g/(kg·d)]和瑞舒伐他汀预处理组[瑞舒伐他汀20 mg/(kg·d)+大量饮酒组50%食用乙醇,5 g/(kg·d)]。喂养4周后制做心肌梗死模型,于术后第3天处死,采用免疫组织化学SP法检测梗死边缘区心肌内微血管密度(MVD),应用ELISA法测定心肌组织中血管内皮生长因子(VEGF)及内皮抑素的表达。结果与空白对照组相比,大量饮酒组MVD明显减少(P<0.01),VEGF表达无明显差异,内皮抑素表达明显增加(P<0.01);与大量饮酒组相比,瑞舒伐他汀预处理组MVD明显升高(P<0.01),VEGF和内皮抑素水平均无明显差异(P>0.05)。结论大量饮酒抑制心肌梗死大鼠缺血心肌的血管新生,可能与增加内皮抑素的表达有关;瑞舒伐他汀能够减轻大量饮酒对缺血心肌血管新生的抑制作用。 Objective To study the effect of heavy drinking on angiogenesis in ischemic rats and the effect of Rosuvas-tatin on angiogenesis after heavy drinking.Methods A total of 24 male SD rats were randomly divided into 3 groups:control group, heavy-drinking group[50%ethanol, 5 g/(kg&#183; d)] and Rosuvastatin pretreated group[Rosuvastatin (20 mg/(kg&#183; d)+50%ethanol, 5 g/(kg&#183; d)].The three groups were given equal volume of ethanol, rosuvastatin and vehicle by gavage respectively for 4 weeks and then myocardial infarction models were constructed.The rats were killed on the 3rd day after myocardial infarction.The microvascular density ( MVD) was observed by immunohistochemical SP method.The vascular endothelial growth factor ( VEGF) and endostatin expressions in the ischemia myocardium tis-sue were tested with ELISA.Results Compared to the control group, the MVD was significantly reduced ( P&lt;0.01) in the heavy-drinking group, the level of endostatin was significantly increased (P&lt;0.01), and there was no difference in the expression of VEGF.MVD in the Rosuvastatin pretreated group was significantly increased compared to that in the heavy-drinking group (P&lt;0.01), but the VEGF and endostatin expressions were not significantly different between the two groups(P&gt;0.05).Conclusion Heavy drinking could inhabit angiogenesis by increasing the expression of en-dostatin after myocardial infarction.Rosuvastatin could reverse the damages of heavy drinking on angiogenesis.
