番茄叶中总黄酮提取工艺及含量测定的研究

摘要 为了优化番茄(Lycopersicon esculentum Mill.)叶总黄酮含量的最佳提取工艺并测定其总黄酮的含量。以总黄酮含量为考察指标,采用正交设计的方法研究提取番茄叶总黄酮的工艺条件,并测定其总黄酮含量。结果表明,影响番茄叶总黄酮提取的主次因素依次为甲醇浓度、溶剂量、超声时间、浸泡时间;建立了番茄叶总黄酮含量的测定方法。其总黄酮的最佳提取工艺为100%甲醇20 mL,浸泡30 min,超声30min;总黄酮的含量测定方法简便可靠。 The extraction technology for total flavonoids were optimized with the content of total flavonoids as test index by orthogonal design. A novel method for determining total flavonoids was developed. The results showed that the factors affecting the total flavonoids extraction were in the order of methanol concentration, solvent amounts, ultrasonic time and soak time. The best extraction technology was as follows: 20 mL 100% methanol, soak time 30 min, ultrasonic time 30 min. The method of determing total flavonoids content was proved to be feasible, effective and simple.
