摘要
以金银花的根、茎、叶为材料,采用组织块法开展内生细菌的分离筛选,并依次借助薄层层析(TLC)、高效液相色谱(HPLC)和液相色谱-质谱联用(LC-MS)对内生细菌的发酵产物进行分析,最终从金银花的叶中获得一株具有产绿原酸能力的细菌B3.菌株B3的绿原酸产量为5.653 mg/L.根据形态特征和16SrDNA序列分析鉴定该内生菌株为Bacillus amyloliquefaciens,并通过构建进化树对B3进行了系统发育分析.B3对几种细菌和真菌具有较强的抑菌作用,其抑菌活性物质主要来自于胞外分泌物.
The endophytic bacteria were isolated from Flos Lonicerae japonicae by the tissue separating method. Fermentation products of isolated bacteria were analyzed by thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC) and liquid chromatography-mass spectrometry (LC- MS) analysis suecessively. As a result, one bacterium named t33 from the leaf of Flos Lonicerae japonicae was found to produce ehlorogenic acid and its yield of ehlorogenic acid reached 5. 653 mg/L. The strain B3 was identified as Bacillus amyloliquefaciens according to the morphological characteristics and 16S rDNA sequence analysis. The phylogenetic analysis of strain B3 was made through cladogram. The strain B3 has better antimicrobial active to several kinds of bacteria and fungi. The antimicrobial active substances of strain B3 were extracellular secreted mainly.
