摘要
目的探讨缺血后处理联合肢体缺血后处理对大鼠局灶性脑缺血再灌注损伤的影响。方法将80只SD大鼠随机分为5组:假手术组(Ⅰ组)、缺血再灌注组(Ⅱ组)、近端缺血后处理组(Ⅲ组)、肢体远隔缺血后处理组(Ⅳ组)、联合处理组(Ⅴ组)。采用线栓法制作局灶性脑缺血再灌注损伤模型。再灌注24h后测定各组大鼠神经行为学评分、脑梗死体积、丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性及肿瘤坏死因子α(TNF-α)水平。结果各组大鼠脑梗死体积比值为0、(55.29±16.34)%、(36.07±14.76)%、(37.23±15.13)%和(21.15±10.92)%;与Ⅰ组相比,其余四组大鼠神经行为学评分、TNF-α水平及MDA含量均明显增高,SOD活性明显降低(均P<0.01);Ⅲ、Ⅳ、Ⅴ组SOD活性高于Ⅱ组,其余指标均低于Ⅱ组(均P<0.05);Ⅲ、Ⅳ组SOD活性低于Ⅴ组,其余指标均高于Ⅴ组(均P<0.05);Ⅲ组与Ⅳ组间各指标差异无统计学意义(均P>0.05)。结论缺血后处理联合肢体缺血后处理能减轻大鼠局灶性脑缺血再灌注损伤,其效果优于单独一种方法。
Objective To investigate the effects of combination of ischemic postconditioning and limb ischemic postconditioning on focal cerebral ischemia-reperfusion injury in rats.Methods A total of 80 SD rats were randomly divided into 5 groups:Group sham operation (Group Ⅰ),Group ischemia reperfusion (Group Ⅱ),Group ischemic postconditioning (Group Ⅲ),Group limb ischemic postconditioning (Group Ⅳ) and Group combined pestconditioning(GroupⅤ).The model of focal cerebral ischemia-reperfusion was induced by the method of thread embolism.The neurologic deficit scores were obtained after 24 h of reperfusion.The rots were decapitated and brains were removed rapidly for determination of the cerebral infarct size,malondialdehyde (MDA) content,superoxide dismutase(SOD) activities and tumor necrosis factor alpha(TNF-α)level.Results The size of cerebral infarct were 0,(55.29 ±16.34)%,(36.07 ± 14.76)%,(37.23 ±15.13)% and (21.15 ± 10.92)% ;Compared with Group I,the scores of neurologic deficit,the level of TNF-α and the content of MDA were significantly increased,while the activities of SOD were significantly decreased in the other 4 groups (all P < 0.05).Compared with group Ⅱ,the activities of SOD were increased,while other indexs decreased in the Groups Ⅲ,Ⅳ and Ⅴ (all P < 0.05).Compared with group Ⅴ,the activities of SOD were decreased,while other indexs increased in the Groups Ⅲ and Ⅳ(all P < 0.05).The indexs were not different among Group Ⅲ and Ⅳ (all P > 0.05).Conclusions The combination of ischemic postconditioning and limb ischemic postconditioning can attenuate the injury of focal cerebral ischemiareperfusion in rats.And the treatment combined displays synergistic effect.
