摘要
目的研究Tim-l,Tim-3及IL-2,IL-4在过敏性紫癜(Henoch-Schonlein purpura,HSP)发病中的作用。方法将28例HSP患者分为单纯组和混合组,采用RT-PCR技术检测两组患者外周血单个核细胞中Tim-1 mRNA和Tim-3 mRNA的表达情况,ELISA法检测血清中IL-2和IL-4的含量,选取健康人20例作为对照组。结果 HSP患者Tim-1 mRNA相对表达量高于正常对照组[RQ值分别为0.974(1.108)和0.760(0.514),P<0.05],而Tim-3 mRNA相对表达量差异无统计学意义[RQ值分别为1.359(1.183)和1.604(1.177),P>0.05];Tim-1 mRNA,Tim-3 mRNA的表达在HSP单纯组和混合组之间差异均无统计学意义(P均>0.05)。HSP患者血清IL-2含量(188.517±12.867)较正常对照组(202.759±20.903)降低,IL-4含量(73.046±8.750)高于正常对照组(62.301±11.232),且HSP患者混合组IL-2含量低于单纯组,IL-4含量高于单纯组,以上差异均有统计学意义(P均<0.05)。结论Tim蛋白可能参与HSP发病,但与疾病严重程度无关;IL-2和IL-4水平可能与疾病严重程度密切相关。
Objective To explore the roles of Tim-l, Tim-3, IL-2and IL-4 in the pathogenesis of HSP. Methods A total of 28 patients with HSP including skin type and mixed type, and 20 normal controls were enrolled in this study. The expression levels of Tim-1 and Tim-3 mRNA were measured by RT-PCR. The serum levels of IL-2 and IL-4 were measured by enzyme-linked immunosorbent assay (ELISA). Results The relative expression level(RQ) of Tim-1 mRNA in HSP patients was higher than the normal[ 0. 974 (1. 108 ) vs 0. 760 (0.514) ,P 〈 0.05 ] while the levels of Tim-3 mRNA were similar in these two groups[ 1. 359( 1. 183 )vs 1. 604 (1. 177 ) ,P 〉 0.051. In HSP patients, the RQ values of Tim-1 and Tim-3 mRNA were no significant difference between the skin type and the mixed type ( P 〉 0.05 ). Compared with control group, the levels of IL-2 in HSP patients were lower( 188. 517 ± 12. 867 vs. 202. 759 ± 20.903, P 〈 0.05 ) and the levels of IL- 4 were higher(73.046 ±8.750 vs. 62. 301 ± 11. 232, P 〈0.05) in HSP patients. Moreover,the levels of IL-2 were lower in the mixed type than the skin type. But the levels of IL-4 were higher in mixed type(P 〈 0.05 ). Conclusion Tim family probably takes part in HPS pathogenesis, but has no impact on the severity of the disease. IL-2 and IL-4 mav be closelv related to the severity of the disease.
