射干麻黄汤加味对哮喘小鼠肺组织过氧化物酶体增殖物激活受体γ表达的影响 被引量:3

摘要 目的探讨射干麻黄汤加味对慢性哮喘小鼠肺组织过氧化物酶体增殖物激活受体γ(PPAR-γ)表达的影响。方法将50只健康清洁级雌性BALB/c小鼠随机分为正常对照组、模型组、地塞米松组、射干麻黄汤组及射干麻黄汤加味组,每组各10只。建立慢性哮喘小鼠模型后,各药物组分别予相应药物干预,正常对照组及模型组予0.9%氯化钠注射液腹腔注射。连续4周后取各组小鼠肺组织,采用免疫组织化学染色(SP法)、逆转录-聚合酶链反应(RT-PCR)技术检测小鼠肺组织内PPAR-γ的数量及转录表达情况。结果与正常对照组比较,模型组小鼠的PPAR-γ的数量及转录表达增加(P<0.05),经地塞米松组处理后PPAR-γ的数量及转录表达有所降低(P<0.05),而射干麻黄汤及射干麻黄汤加味组均有所增加(P<0.05),射干麻黄汤加味组尤为明显(P<0.05)。结论射干麻黄汤加味可抑制BALB/c哮喘小鼠哮喘的发生,其机制有可能是通过升高PPAR-γ的数量及转录表达而实现的。 Objective To explore the effect of Shegan Mahuang decoction on the expression of peroxisome pro -liferator-activated receptor γ( PPAR-γ) in lung tissue of Asthmatic mice .Methods 50 healthy clean female BALB /c mice were randomly divided into normal control group , model group , Dexamethasone group , Shegan Mahuang decoction group and improved Shegan Mahuang decoction group , 10 mice in each group .After establishing a mouse model of chronic asthma , corresponding to each drug group were drug intervention , the normal control group and model group were 0.9%sodium chloride injection by intraperitoneal injection .After four consecutive weeks mice lung tissue of each group were taken , the number and the expression transcription of PPAR -γin mouse lung tissue were detected using immunohistochemical staining (SP), RT-PCR technique.Results The number and the ex-pression transcription of PPAR-γin mouse lung tissue in model group were increased as compared with those in nor-mal control group (P〈0.05).The number and the expression transcription of PPAR -γwere decreased after treat-ment of Dexamethasone (P〈0.05).Those in Shegan Mahuang decoction group and improved Shegan Mahuang de-coction group were increased (P〈0.05).The increase in improved Shegan Mahuang decoction group was obvious (P〈0.05).Conclusion Shegan Mahuang decoction can inhibit incidence of asthma in BALB /c mice with asth-ma, the mechanism may be achieved by increasing the number of PPAR -γexpression and transcription .
