腺苷预处理对糖氧剥夺损伤星形胶质细胞的保护作用

摘要 目的:观察糖氧剥夺对星形胶质细胞的影响及腺苷的保护作用。方法:取体外培养第三代或第四代星形胶质细胞,传代至96孔板或24孔板内,细胞贴壁并长出突起后,随机分为三组:正常对照组(normal control group)、糖氧剥夺组(OGD group)、腺苷预处理组(ADO group),进行糖氧剥夺/复氧处理。观察糖氧剥夺8h复氧糖24h后星形胶质细胞形态、细胞活性。结果:糖氧剥夺复氧糖24h后,正常对照组星形胶质细胞生长状态良好;OGD模型组星形胶质细胞胞体水肿变圆,细胞形态由不规则变成梭形,突起明显变短或消失;ADO组细胞水肿较轻。OGD模型组星形胶质细胞活力下降,OD值明显低于正常对照组(P〈0.01);ADO组细胞活力显著高于OGD模型组,表明ADO能明显改善细胞活力(P〈0.01)。结论:糖氧剥夺可引起星形胶质细胞严重损伤,星形胶质细胞形态、活性与缺氧缺糖呈对应变化,腺苷能减少糖氧剥夺后星形胶质细胞形态学改变及细胞凋亡,提示腺苷对糖氧剥夺的星形胶质细胞有一定的保护作用。 Objective To investigate the effect of oxygen--glucose deprivation and protective effect of adenosine on astrocytes. Methods Astrocytes were cultured for third or fourth generation in vitro, and then passaged at 96 or 24 well plate. When the astrocytes attaching to the wall and growing with prominences, they were randomly divided into 3 groups, normal control group, oxygen-glucose deprivation (OGD) group and adenosine preconditioning (ADO) group. The cells were taken OGD/reoxygenation treatment. The morphology and activity of astrocyte cells were observed at 8 hours after OGD and 24 hours after reoxygenation. Results At 24 hours after OGD/reoxygenation treatment, the astrocyte cells were with normal morphology in normal control group; the soma of astrocyte cells became edema and round in OGD group, the cell morphology changed to fusiformis from irregular, and prominence obviously became short or disappeared; the edema was lighter in ADO group. Activity of astrocyte cells decreased in OGD group, and OD value was significantly lower than that in normal control group (P〈0.01). Cell activity was higher in ADO group than that in OGD group, which indicated that ADO can obviously improved cell activity (P〈0. 01). Conclusions Astrocytes are injured seriously after OGD; morphology and cell activity of astrocytes take change in accordance with oxygen deficit and glucose deprivation. Adenosine can decrease naorphnlogical changes and cell apoptosis of astrocyte cells after OGD. we can make conclusion that adenosine has influence on astrocytes after OGD.
