摘要
为明确彩色马铃薯优良株系16、株系19、株系26、株系31及亲本拉塞特-1和黑美人在DNA水平上的遗传差异程度,对其进行了SSR分析.利用筛选出的10对SSR适宜引物,扩增得到89个多态性位点,多态性位点百分率为92.1%.试验建立了4个彩薯优良株系及其亲本的SSR指纹图,为其在DNA水平上鉴别及新品种选育提供了依据.6个供试材料间的遗传距离(GD)变幅为0.3506~0.754 1,以GD值0.42为基准将其分为3类:母本拉塞特-1、株系16、株系19和株系26为一类,株系31和父本黑美人单独为一类.
In order to determine the genetic differences of promising color potato line 16, line 19, line 26, line 31 and their parents Russet-01and Heimeiren at DNA level, the 6 materials were analyzed by SSR maker technology. Use the screened 10 SSR suitable primer pairs to amplify 89 polymorphic loci, accounting for 92.1% of all the amplified loci. The SSR fingerprint of the four colorful potato strains and their parents was established, which provided a basis for the identification about promising lines and breeding of new cuhivars at DNA level. The genetic distance (GD) of six tested materials was ranged from 0.092 2 to 0. 754 7. The materials were classified into three categories on the basis GD value of 0.42 :mother Russet-01, new line 16,19, and 26 as a category, and new line 31 and farther Heimeiren as a category.
