蜗牛多肽混合物对过氧化氢诱导的SH-SY5Y细胞PCNA表达的影响

Effect of Snail Polypeptide Mixture on Expression of PCNA in Injured SH-SY5Y Cells Induced by Hydrogen Peroxide

摘要 采用H2O2诱导人神经母细胞瘤细胞株(SH-SY5Y)细胞损伤,MTT法测定细胞存活率,不同浓度蜗牛多肽混合物(SPM)处理后,Hoechst染色检测细胞凋亡,细胞免疫化学技术和Western blot技术检测细胞PCNA的表达。结果发现1.54 mmol/L H2O2可诱导SH-SY5Y细胞凋亡,PCNA的表达明显降低。经用39 mg/L(SPM-L组)和156 mg/L(SPM-H组)浓度的SPM处理后,SH-SY5Y细胞凋亡明显减少(H2O2VS.SPM-L:58.39±8.67%VS.44.06±4.35%,P<0.05;H2O2VS.SPM-H:58.39±8.67%VS.32.45±9.44%,P<0.01),同时PCNA的表达呈浓度依赖性升高。提示SPM可抑制H2O2诱导的SH-SY5Y细胞凋亡,其作用机制可能与其增加细胞PCNA的表达有关。 In this study,hydrogen peroxide (H2O2 ) was used to induce SH-SYSY cells damage. The viability of SH- SY5Y ceils was then measured with MTT assay. Ceil apoptosis was detected with Hoechst staining. The expression of PCNA was determined with immunocytochemistry and western blotting. The results showed that 1.54 mmol/L H202 can significantly induce apoptosis, and decrease expression of PCNA in SH-SY5Y ceils. After treatment with 39 mg/L (SPM-L) and 156 mg/L (SPM-H) of snail polypeptide mixture (SPM), H202-induced apeptosis ceils were attenuated (H2O2 VS. SPM-L:58.39 ±8.67% VS. 44.06 ±4.35% ,P 〈0.05;H2O2 VS. SPM-H:58.39±8.67% VS. 32.45±9.44% ,P 〈 0.01 ) ,while there was a concentration-dependent increase of PCNA expression in the SPM groups. These results suggested that the SPM can protect against H2O2 -induced SH-SYSY cells apoptosis,which may be correlated with up-regu- lated PCNA expression.
