摘要 目的探讨胶质细胞源性神经营养因子(GDNF)诱导骨髓间充质干细胞(BMSCs)促周围神经再生。方法建立大鼠坐骨神经损伤模型,实验分为2组:对照组为未经诱导BMSCs组,实验组为GDNF基因诱导后BMSCs组。每组20只SD大鼠。①术后4周和8周观察坐骨神经指数和腓肠肌湿质量恢复率。②术后8周测量再生神经纤维轴突直径、髓鞘厚度及有髓神经纤维计数。③术后4周腓肠肌肌细胞行DAPI染色。④术后8周坐骨神经扫面电镜观察神经丝再生交联情况。结果经GDNF基因诱导BMSCs组各方面检测指标均较对照组为好,其中诱导后实验组扫描电镜下观察周围神经再生更加明显。结论 GDNF基因诱导的间充质干细胞对周围神经再生有着显著的疗效。 Objective To clarify the role of Glial cell derived neurotrophic factor (GDNF)inducing bone marrow mesenchymal stem cells in promoting regeneration of peripheral nerve.Methods Sciatic nerve injury models of rats were established,Experiment was divided into two groups:experimental group was BMSCs group induced by GDNF gene,and control group was BMSCs group un-induced by GDNF gene ,each group had 20 SD rats.① Observe index of sciatic nerve and gastrocnemius wet quality recovery rate after 4 weeks and 8 weeks.② Measured axon diameter ,thick-ness of myelin sheath and myelinated nerve fiber numbers of regeneration nerve fiber 8 weeks postoperative.③The gas-trocnemius muscle cells were stained with DAPI after 4 weeks postoperative.④ Neurofilament regeneration crosslink-ing of the sciatic nerve were obsvered by sweep electron microscope after 8 weeks.Results Test index of the BMSCs group induced by the GDNF gene is better than those of the control group,peripheral nerve regeneration of experimen-tal group observed by scanning electron microscopy was better than those of control group.Conclusion bone marrow mesenchymal stem cells induced by GDNF gene have significant effection for the regeneration of peripheral nerve.
