摘要
为研究人工合成的过氧化物酶体增值物激活受体α特异性受体激活剂(WY14643)对附植前小鼠胚胎体外发育的影响,以及WY14643对附植前小鼠胚胎内部活性氧的影响,通过H2O2诱导建立胚胎氧化损伤模型,添加不同浓度WY14643对胚胎进行体外培养,利用2’7’二氯荧光素二乙酸盐(DCHFDA)测定胚胎内部H2O2含量。结果表明:不同浓度WY14643处理2 h后,0.1μmol/L WY14643处理组4-细胞发育率、囊胚率显著高于其他各组(P<0.05);经0.1μmol/L WY14643和H2O2联合处理后,其胚胎发育率显著高于单独H2O2处理组和对照组(P<0.05);DCHFDA荧光检测发现,WY14643添加组胚胎内部各时期H2O2水平显著低于其他各组(P<0.05),并且胚胎内部H2O2呈一定的变化规律。说明适宜浓度的WY14643能够增加体外发育胚胎的发育率,减少胚胎内部活性氧水平,并且胚胎内部H2O2水平在4-细胞时期达到最高,囊胚时期显著下降。
The influence of WY14643, a synthetic specific receptor activator of PPAR-α(peroxisome proliferator ac-tivated receptor α), on the development of preimplantation mouse embryos were studied by establishing a H2 O2 -induced ox-idative stress model in mouse embryo cultured in vitro. H2 O2 levels in embryos were measured by DCHFDA (dichloro fluo-rescein diacetate salt). After 2-h treatment with different concentrations of WY14643, the concentration of 0.1 μmol/ Lshowed better effects on 4-cell growth rate and blastocyst growth rate of mouse embryo than other concentrations. The embyogrowth rate co-treated by 0.1 mol/ L WY14643 and H2O2 was higher than those in solo H2O2 treatment and control group.The H2O2 levels of mouse embryos in WY14643 applied group at each developmental stage were much lower than those inother groups. It is concluded that approapriate concentration of WY14643 can increase the growth rate of embryo cultured invitro and decrease H2O2 level which peaked at 4-cell stage and declined rapidly at blastocyst stage.
