摘要
为了用响应曲面法优化微波辅助提取苦荞籽总黄酮的工艺,试验选用微波辅助法提取苦荞籽总黄酮,并选取乙醇浓度、料液比、提取时间、微波预处理时间4个因素进行单因素试验,在此基础上以4个主要因素(乙醇浓度、料液比、提取时间、微波时间)为自变量,提取率为响应值,采用Box-Behnken试验设计和响应面分析法,建立数学模型。结果表明:确定苦荞仔黄酮提取的最佳工艺条件为乙醇浓度58.25%,料液比27.97∶1,提取时间2.38 h,微波时间107.48 s;在此条件下,苦荞仔总黄酮的得率5.05%。说明由响应曲面法得到的优化工艺参数比较准确,具有实用意义。
To optimize the microwave assisted extraction process of flavonoids from tartary buckwheat seeds using response surface methodology (RSM). The microwave - assisted extraction was used to extract total flavonoids (TF) from tartary buckwheat seeds, and the following four fac- tors were selected to be used for a single factor test, including the ethanol concentration, liquid - material ratio, extraction time and microwave pretreatment time ; the Box - Behnken experimental design and RSM were used to build a mathematical model with the extraction rate as the re- sponse value based on the four factors as independent variables, including the ethanol concentration, liquid - material ratio, extraction time and microwave time. The results showed that the optimal conditions for the extraction of flavonoids from tartary buckwheat seeds were determined as follows : the ethanol concentration of 58.25 %, the liquid - material ratio of 27.97:1, the extraction time of 2.38 h, and the microwave time of 107.48 s. Under these conditions, the obtained extraction yield of total flavonoids from tartary buckwheat seeds was 5.05%. The results indicate the technological parameters optimized by RSM are more accurate,which have practical significance for the extraction of total flavonoids from tar- tary buckwheat seeds.
