摘要
目的探讨细胞毒性 T淋巴细胞相关抗原 4(CTL A- 4)基因外显子 1的 49位点 A/G多态性与自身免疫性甲状腺病 (AITDs)的相关性。方法采用多聚酶链反应限制性片段长度多态性 (PCR- RFL P)技术分析 12 2例自身免疫甲状腺病患者 ,其中 Graves’病 (GD) 87例 ,桥本甲状腺炎 (HT) 35例 ,84例健康对照的 CTL A- 4基因外显子 1的 49位点基因型。采用 EL ISA技术检测 AITDs患者甲状腺功能 ,间接免疫荧光法检测甲状腺球蛋白抗体 (TGAb)和甲状腺抗过氧化物酶抗体 (TPO Ab)。结果 AITDs患者 CTL A- 4/G49等位基因频率显著高于对照组 (P<0 .0 0 0 1,其中 GD组 P<0 .0 0 0 1,HT组 P<0 .0 1) ;GD、HT组按性别分层分析后发现 CTL A- 4/G49等位基因在不同性别的分布均无显著性差异。结论 CTL A- 4基因外显子 1G49等位基因与 AITDs显著相关 ;GD、HT患者
ObjectiveTo investigate the association of gene polymorphism of cytotoxic T lymphocyte associated antigen 4(CTLA 4) with autoimmune thyroid diseases MethodsThe A/G transition polymorphism at position 49(exon 1,codon 17) of the CTLA 4 gene was determined by polymerase chain reaction restriction fragment length polymorphism (PCR RFLP)method in 122 autoimmune thyroid diseases patients which included 87 Graves’ disease (GD) patients and 35 Hashimoto's thyroiditis(HT) patients, as well as 84 control subjects We detected their thyroid function by ELISA technique, and the thyroid autoimmune antibodies (TGAb,TPOAb) by indirect immunofluorescent technique ResultsThe strong association of the CTLA 4/G 49 allele with AITDs was seen in our study(66 4% vs 36 9% P <0 0001) The G allele in GD patients was significantly increased compared with control subjects(69 5% vs 36 9%, P <0 0001) In HT patients, the frequency of G allele was also higher than control subjects(58 6% vs 36 9%, P <0 01), and there was no significant difference between HT and GD groups When GD and HT subjects were stratified with respect to sex, neither female nor male patients demonstrated evident association of G 49 allele with gender ConclusionsThe polymorphism of CTLA 4 gene (exon 1 condon 17 position 49)confers susceptibility to AITDs This association is independent of sex
