姜黄素诱导骨髓间充质干细胞分化成肝细胞 被引量:9

摘要 目的研究姜黄素诱导大鼠骨髓间充质干细胞分化为肝细胞的作用。方法贴壁筛选法体外培养骨髓间充质干细胞,流式细胞仪检测其表面抗原。分组诱导培养14 d:1组(空白对照组),2组(10 ng/mL成纤维细胞生长因子-4,FGF-4),3组(10 ng/mL FGF-4+20 ng/mL肝细胞生长因子,HGF),4组(10 ng/mL FGF-4+5μmol/L姜黄素),5组(20μmol/L姜黄素)。观察细胞形态学改变;利用western-blotting分析肝特异性蛋白HNF-3β的表达。结果大鼠骨髓间充质干细胞细胞表面抗原CD34、CD14、CD45阴性表达,CD44、CD73、CD90表达阳性;药物诱导后光镜下观察到骨髓间充质干细胞细胞由梭形渐变成圆形肝细胞;诱导分化后的细胞表达肝特异性蛋白HNF-3β。结论姜黄素具有促进骨髓间充质干细胞分化为肝细胞的作用。 AIM To investigate whether curcumin can induce differentiation of rat bone mesenchymal stem cells (rBMSCs) into hepatocytes in vitro. MWTIJODS rBMSCs were isolated from adult SD rats, and then were cultured and cloned by density gradient method and adhesive cultivation. Their cell surface antigens were detected with flow cytometer, rBMSCs obtained were randomly assigned to five groups: group 1 (control) , group 2 (in- duced by 10 ng/mL FGF-4), group 3 (induced by 10 ng/mL FGF-4 combined with 5 μmol/L curcumin), group 4 (induced by 10 ng/mL FGF-4 combined with 20 ng/mL HGF), and group 5 (induced by 20 μmol/L curcu- min). After induction, hepatocytes cell percentage was determined, and the expression of specific proteins of HNF-313 was analyzed. RESULTS CD44, CD73 and CD90 expression of rBMSCs was positive while that of CD34, CD14 and CD45 was negative, indicating the successful culture of rBMSCs. After induction with curcurain, rBMSCs showed morphological variation. The expression levels of HNF-3β in group 2, 3, and 4 were significantly higher than those in the group 1. CONCLUSION Curcumin can induce rBMSCs to differentiate into hepa- tocytes cells in vitro.
