摘要
目的 探讨兔眼玻璃体腔内注射不同剂量的白蒺藜皂苷(gross saponins from tribulus terrestris L,GSTT)对视网膜的毒性作用.方法 动物实验研究.对2012年6~12月在深圳大学医学院实验动物中心.应用健康新西兰大白兔24只,按照随机数字表法随机分为4组,每组6只(6只眼).实验组按照注射剂量分为3组:A组玻璃体腔注射低剂量(1 mg/kg) GSTT,B组玻璃体腔注射中剂量(3 mg/kg) GSTT,C组玻璃体腔注射高剂量(5 mg/kg) GSTT.对照组(D组):玻璃体腔内注射PBS0.1 ml.用裂隙灯显微镜、直接检眼镜、眼部B超活体观察兔眼眼前节和眼底变化情况,于用药后4周处死兔,取视网膜行光学显微镜和透射电镜观察.结果 (1)对照组和实验组兔眼前后节未见明显异常.(2)对照组和实验组光学显微镜观察视网膜组织形态和透射电镜观察视网膜超微结构均未见明显异常改变.结论 兔眼玻璃体腔注射不同浓度的(1、3和5 mg/kg)白蒺藜皂苷,对兔视网膜无明显毒副反应.
Objective To evaluate the retinal toxieity of intravitreal gross saponins from tribulus terrestris L (GSTT) in normal rabbit retina.Methods The experiment was done in laboratory animal center of Shenzhen university medical school on January to December 2012.Twenty four New Zealand albino rabbits randomly divided into four groups according to random number table.Three experimental groups named group A,B,C,and different concentrations (1mg/kg,3mg/kg,5mg/kg,respectively) of GSTT in 0.1ml were injected intravitreously.The other group named group D as a control,and accepted intravitreal injection PBS 0.1ml.With slitlamp,ophthalmoscope,and B ultrasound to observe the anterior segment and fundus changes in rabbit eyes.After 4 weeks,microstructure of retina was observed under light microscopy and transmission microscopy.Results There were no apparent forepart and fundus changes of eyes in each group.By the light microscopy and transmission microscopy observation,there were no obvious abnormal changes of retinal ultrastructure and retinal layers histologic morphology in each group.Conclusions There are no retinal toxieity of intravitreal injection of different concentration GSTT on normal rabbit retina.
