摘要 目的研究探讨自体组织工程化软骨在动物自体内最佳形成时间 ,为临床应用提供理论研究和参数。 方法取猪耳廓软骨细胞 ,与可注射性生物材料PluronicF12 7混合形成浓度为 5 0× 10 6 /ml细胞—生物材料复合物 ,接种于猪自体腹外侧壁真皮下 ,分别在接种后第 1、3、6、9、15周取材 ,从组织学、生化组成对再生软骨组织进行评价。 结果第六周形成的软骨组织完全成熟 ,与正常软骨组织有相似的组织学特性。而第 1、3周大部分为未成熟的软骨细胞。各不同时间再生软骨组织中氨基糖氨多糖 (GAG)占组织干重的平均百分含量在 7.0~ 9.0之间。第 6周形成软骨组织中Ⅱ型胶原含量比第 1、3周形成软骨组织中Ⅱ型胶原含量高 ,和正常耳软骨含量接近。 结论利用组织工程技术可在具有免疫功能自体动物内形成软骨组织 ,确定软骨组织形成的最佳时间为接种后第 6周。 Objective The study is to investigate the best formation time of the tissue engineered autologous cartilage and provide theoretical basis and parameters for clinical use. Chondrocytes isolated from the mini swines ears were mixed with injectable biocompatible matrix (Pluronic F127) to generate cell suspensions with densities of 50×10 6/ml. The chondrocyte-polymer compounds were injected into the subcutaneous layer of the swine abdomen where the cells had been isolated. Specimens were harvested and analyzed by histology for the content of glycosaminoglycan and type Ⅱ collagen in 1,3,6,9,15 weeks after transplantation. Histologic analysis of the neocartilage showed matured chondrocytes enclosed in the lacuna with similar histological characteristics to the natural cartilage in 6 weeks. Most of newborn cartilage are immature chondrocytes, and areas of the cartilage are separated by remnant polymers in 1,3 weeks. The glycosaminoglycan content ranged from 7.0 to 9.0 percent for all samples; normal auricular cartilage controls had a content of 9.2 percent. Western Blot demonstrated the presence of type Ⅱ at a higher level than that formed in 1 and 3 weeks. The experiments demonstrated that using tissue engineering skills we can generate an autologous cartilage with similar histological characteristics to the natural cartilage on animals with the normal immune system. The optimal formation time of tissue engineered autologous cartilage was at 6 weeks according to the analysis of histology on the content of glycosaminoglycan and type Ⅱ collagen.
