摘要
目的:研究药对巴戟天-杜仲总多糖对体外培养的大鼠软骨细胞增殖活性的影响。方法:①用水提醇沉法、Sevag法除蛋白提取纯化获得其总多糖,苯酚-硫酸比色法测定其总多糖含量;②MTT法分别检测体外培养大鼠软骨细胞在800,400,200,100,50μg·mL-1总多糖DMEM溶液中24,48,72 h的增殖能力。结果:①葡萄糖标准品线性回归方程为y=0.0100x-0.0018(r=0.9996),葡萄糖标准品在10~100μg·mL-1范围内线性关系良好,平均加样回收率为99.59%,RSD为0.40%;多糖含量为43.88%。②巴戟天-杜仲总多糖对软骨细胞具有一定的促增殖作用,当作用时间为48 h、多糖浓度为400μg·mL-1时,促增殖作用最强。结论:①苯酚-硫酸比色法可作为药对巴戟天-杜仲总多糖的含量测定方法;②巴戟天-杜仲总多糖对软骨细胞的促增殖作用具有一定的时效量效关系。
Objective:To study the effects of polysaccharide in couplet drugs of Bajitian(Radix Morindae Officinalis)and Duzhong(Cortex Eucommiae)on the proliferation of rat chondrocytes cultured in vitro.Methods: ① The total polysaccharide was extracted and purified by the water extraction and alcohol pre-cipitation and the Sevag method,measuring its content by Phenol-sulfuric acid colorimetric method; ② By the MTT method,the reproductive activity of rat chondrocytes was detected in the polysaccharide DMEM solution of 800,400,200,100,50 μg·mL-1 for 24,48,72 hs.Results: ① The standard glucose linear regression equation was y=0.010 0x-0.001 8 (r=0.999 6).The linear relationship of standard glucose was good in 10~100 μg·mL-1, the average recovery being 99.59%,RSD being 0.40%and the content of polysaccharide being 43.88%. ② The total polysaccharides of Bajitian(Radix Morindae Ofifcinalis)and Duzhong(Cortex Eucommiae)could pro-mote the reproductive activity of rat chondrocytes.The promoting function was strongest when the concentration of polysaccharide was 400 μg·mL-1 for 48 hs.Conclusion:①The Phenol-sulfuric acid colorimetric method can be used to measure the content of polysaccharide in couplet drugs of Bajitian(Radix Morindae Ofifcinalis)and Duzhong(Cortex Eucommiae);②The function of polysaccharide in promoting the reproductive activity of rat chondrocytes is related to a certain time,quantity and effectiveness.
