摘要
目的探讨针刺对过敏性哮喘(哮喘)大鼠肺肠组织表面蛋白A(SP-A)mRNA表达的影响。方法将40只SD大鼠随机分为空白组、哮喘模型组、针刺肺组、针刺肠组、针刺肺肠组,每组各8只。除空白组外,余4组采用腹腔注射卵蛋白致敏,2周后尾静脉注射卵蛋白激发哮喘发作进行造模。空白组、哮喘模型组不针刺,针刺肺组针刺尺泽、孔最、列缺、肺俞穴,针刺肠组针刺曲池、合谷、天枢、上巨虚穴,针刺肺肠组针刺孔最、列缺、曲池、天枢穴。检测并比较各组大鼠针刺后肺肠等组织中SP-A mRNA的表达。结果哮喘模型组肺、大肠及小肠组织中SP-A mRNA的表达均较空白组高(P<0.01);针刺肺组、针刺肠组、针刺肺肠组的肺、大肠及针刺肺肠组小肠组织中SP-A mRNA的表达均较哮喘模型组高(P<0.05);针刺肺组、针刺肠组的小肠组织中SP-A mRNA的表达与哮喘模型组比较无明显差异(P均>0.05)。结论哮喘大鼠肺肠组织SP-A mRNA表达均发生异常,其中肺与大肠生物学相关性最为密切。
Objective To explore the influence of acupuncture on the expression of pulmonary surfactant protein( SPA) mRNA in the lung and intestine tissues of rats with allergic asthma. Methods A total of 40 SD rats were randomly divided into control group( n = 8),allergic asthma model group( n = 8),the lung acupuncture group( n = 8),the intestine acupuncture group( n = 8) and the lung and intestine acupuncture group( n = 8). Except in the control group,rats in the rest four groups received egg album by intraperitoneal injection to induce allergy,and the models of allergic asthma were established by egg albumin of intravenous injection in vena caudalis. Rats in control and allergic asthma model groups were not treated. Rats in lung acupuncture group received acupuncture treatment in acu-points of Chize( LU11),Kongzui( LU6),Lieque( LU7) and Feishu( BL13); rats in intestine acupuncture group received acupuncture treatment in acu-points of Quchi( LI11),Hegu( LI4),Tianshu( ST25) and Shangjuxu( ST37); rats in the lung and intestine acupuncture group received acupuncture treatment in acupoints of Kongzui( LU6),Lieque( LU7),Quchi( LI11) and Tianshu( ST25). The SP-A mRNA expressions were detected and compared in the five groups after the treatment. Results The expressions of SP-A mRNA in the tissues of lung,large intestine and small intestine in the allergic asthma model groups were significant higher than those in control group( P 〈0. 01); the expressions of SP-A mRNA in the tissues of lung and large intestine in the lung acupuncture,the intestine acupuncture and the lung and intestine acupuncture groups and small intestine tissues in the lung and intestine acupuncture group were significantly higher than those in the allergic asthma model group( P 〈0. 05); the differences in expressions of SP-A mRNA in small intestine tissues between the lung acupuncture and the intestine acupuncture groups with allergic asthma model group showed no statistical significances( P 〈0. 05). Conclusion The SP-A mRNA expressions in the lung and intestine tissues of rats with allergic asthma are of bnormality,and the lung has significant biological relationship with the large intestine.
