摘要
目的比较苦豆子总碱及8种生物碱单体,筛选出具有抗癌活性的药物。方法采用MTT方法测定苦豆子总碱及8种生物碱单体对肝癌、胃癌、结肠癌细胞株的增殖活性影响;Hochest33342-PI双染观察细胞形态学变化,流式细胞仪检测细胞周期及细胞凋亡。结果苦豆子总碱及8种生物碱对消化道癌细胞具有不同程度的增殖抑制作用,其作用呈时间-剂量依赖性,以苦豆碱的抑制作用最强。苦豆碱促进了细胞凋亡,并使细胞阻滞在G2期。结论苦豆子总碱及8种生物碱单体都具有一定的抗癌活性,以苦豆碱的活性最强,为进一步开发抗癌新药提供实验依据。
Objective To screen drugs with anticancer activity by comparing the total alkaloid of Sophora alopecuroides and eight kinds of alkaloid monomer. Methods Determination of total alkali and eight kinds of alkaloids of Sophora alopecuroides monomer for liver cancer, stomach cancer, colon cancer cell line proliferation activity by MTT ;Hochest 33342-PI double dye to watch cell morphology, flow cytometry instrument to detect the cell cycle and cell apoptosis. Results The total alkaloid of Sophora alopecu- roides and eight kinds of alkaloids of digestive tract cancer ceils with different degree of proliferation inhibition ,its role in time - dose dependent. Aloperine has the strongest inhibitory effect. Aloperine promotes the apoptosis and the cell block in G2 phase. Conclusion The total alkaloid of Sophora alopecuroides and eight kinds of alkaloid monomer have anti - cancer activity. Aloperine has the strongest inhibitory effect to provide experiment basis for further screening of anticancer drugs.
