基于正交设计比较色差变化优化植物浸制标本保绿工艺

Optimization of Keeping Green Technology for Plant Immersion Preparations by Orthogonal Design through Color Difference Comparison

摘要 目的:以正交设计的方法优化7种植物叶片的保绿工艺。方法:采用色彩色差计测定7种植物叶片保绿前后平均△E*值作为考察指标,正交设计优化7种植物叶片最佳的保绿工艺。结果:结合正交设计结果及实际情况杀生固定液的配方组成为:1%硫酸铜、1%甲醛、20%乙醇、10%冰醋酸,杀生固定4 d为宜。结论:优选得到的工艺稳定、可行,可作为绿色植物的保绿工艺。 Objective:To optimize the keeping green technology for leaf immersion preparations of 7 species of plants by orthogonal design.Methods:The optimum condition of keeping green was screened by orthogonal design with the average of/△ E* of the leaves of 7 species before and after keeping green as the evaluation index using a color-difference meter.Results:Considering the results of the orthogonal design and actual situation,the optimum keeping green condition was composed of 1% copper sulfate,1% formaldehyde,20% ethano and 10% acetic acid,and the killing time was 4 days.Conclusion:The optimized condition is stable and feasible,and the keeping green technology can be used in keeping green for green plants.
