摘要
本实验建立了高效液相色谱法(HPLC)测定饲料中癸氧喹酯含量的方法。该方法采用C18色谱柱(250 mm×4.6 mm,粒径5μm),以0.025 mol/L氯化钙溶液+甲醇(15∶85,V/V)为流动相,激发波长为326 nm,发射波长为384 nm,流速为1.0 mL/min,外标法定量。结果表明,该方法最低检测限为0.5 mg/kg,癸氧喹酯的浓度为0.02~2.0 mg/L时,峰面积与浓度线性关系良好(r=0.99994),平均回收率在80%以上,变异系数小于10%,稳定性、重现性好,可用于饲料中癸氧喹酯的含量测定。
This experiment was conducted to study the method for high performance liquid chromatographic (HPLC) for determination of decoquinate in feeds by HPLC method.A C18 column (250 mm×4.6 mm,5μm)was used with the mobile phase of 0.025 mol/L calcium chloride-methanol (15∶85,V/V)at the excitation wavelength of 326 nm and emission wavelength of 384 nm with a flow rate of 1.0 mL/min.The quantification was carried out by the external standard method. The limit of quantitation(LOQ)was 0.5 mg/kg.The calibration curves had good linear within 0.02 - 2.0 mg/L(r=0.99994). The average recovery was above 80%.The variation coefficient was less than 10%.The results showed that the method had advantages of good stability and reproducibility,and could be used for the determination of decoquinate in feed.
