摘要
以半饱和荧光萃取实验及竞争性实验探索仲辛基苯氧乙酸(CA-12)/磺化煤油萃取L-Lys的化学作用力。将L-Lys游离氨基用OPA进行半饱和荧光标记后再用CA-12萃取的实验结果指出:在游离氨基/OPA摩尔比低于2∶1区间,萃取相荧光强度随着OPA增加而上升并在游离氨基/OPA摩尔比等于2∶1时达到最大值;当游离氨基/OPA摩尔比大于2∶1,萃取相荧光强度则随着OPA增加而下降并在游离氨基/OPA摩尔比等于1∶1时降至最低;由此证明CA-12通过与L-Lys形成静电相互作用实现萃取。氨基酸、葡萄糖与L-Lys竞争实验结果指出,葡萄糖不影响CA-12/磺化煤油萃取L-Lys,但氨基酸的竞争作用影响CA-12对L-Lys的萃取效率。基于中性氨基酸L-Ala浓度对CA-12/磺化煤油萃取L-lys的贡献,L-Arg浓度的影响为[L-Ala]+[胍基-NH3+],对CA-12萃取L-lys的竞争作用最大;而L-Asp浓度的影响为[L-Ala]-[β-COO-],对CA-12萃取L-lys的竞争作用最小。
The chemical interaction of L-lysine(L-Lys)extracted from the sec-octyphenoxy acetic acid(CA-12)and sulfonated kero-sene was explored and analyzed by the extraction of half-saturated fluorescence and competition experiments. The experiment result of the L-Lys amino labeled by half-saturated fluorescence before extracting with CA-12 showed that the fluorescence intensity of ex-traction phase increases with the addition of OPA when the molar ratio of free amino group and OPA was below 2∶1 and a peak oc-curs at ratio of 2∶1. When the ratio increases continuously and beyond 2∶1,the fluorescence intensity will go down and be minimum at the ratio of 1∶1. Therefore it was proved that the extraction was electrostatic interaction between CA-12 and L-Lys. It was proved that by the competing experiment that glucose doesn’t effect extracting L-Lys of CA-12/sulfonated kerosene,but amino acids had competition function. As the concentration of neutral amino acid L-Ala contributes to the L-Lys extracted from CA-12 and sulfonated kerosene,the concentration of L-Arg influenced extracting L-Lys by CA-12 and the largest competition was equal[ L-Ala]+[ guanidi-no-NH3+] ,while the least competition of L-Asp concentration was equal[ L-Ala]-[β-COO-] .
