摘要
目的研究三种慢性心力衰竭模型大鼠心功能和肿瘤坏死因子α(TNF-α)的变化。方法 60只SD大鼠随机分为冠状动脉结扎组(MI组,12只)、腹主动脉缩窄组(COA组,12只)、腹腔注射异丙肾上腺素(ISO)组(ISO组,12只)及其各自对应的假手术组(sMI、sCOA、sISO组,每组8只)。术后8周,在体测定心功能,取心脏计算心重指数,ELISA法检测心肌组织TNF-α水平。结果 MI、COA、ISO组术后8周存活率分别为75.00%、66.67%、58.33%。与对应假手术组相比,MI组左心室重/心重(LVW/HW)下降、左心室重/体重(LVW/BW)上升,COA组和ISO组LVW/HW、LVW/BW均上升,MI组和ISO组LVW/HW均低于COA组(P<0.05);MI、COA、ISO组左心室收缩压(LVSP)、收缩期左心室内压上升的最大变化速率(+dp/dtmax)、舒张期左心室内压下降的最大变化速率(-dp/dtmax)均下降,左心室舒张末压(LVEDP)上升;三组TNF-α水平均升高,以COA组上升幅度最大(P<0.05)。结论 COA组存活率居中,但心力衰竭表现最为明显。三种心力衰竭模型各有优缺点,应根据具体实验要求和目的选择合适的实验动物模型。
Objective To investigate the changes of cardiac function and TNF-α expression in three different rat models with chronic heart failure(CHF). Methods Sixty SD rats were randomly divided into six groups of MI(coronary artery ligature model, 12 rats), COA(coarctation of abdominal aorta model, 12 rats), ISO ( isoproterenol injection model, 12 rats) and their corresponding sham- operated groups of sMI, sCOA and sISO with 8 rats each. Eight weeks after operation, the cardiac function was observed, and the ratios of left ventricular weight to heart weight (LVW/HW) and left ventricular weight to body weight (LVW/BW) were calculated. The TNF-α expression was detected by ELISA. Results The survival rates of groups of MI, COA and ISO were 75.00%, 66. 67% and 58.33% after eight weeks. Compared with the corresponding sham-operated groups, LVW/HW was decreased and LVW/BW was increased in group MI, and both LVW/HW and LVW/BW were increased in groups of COA and ISO. Besides, left ventricular systolic pressure(LVSP), maximal rate of pressure increase in the contraction phase(q-dp/dtmax) and maximal rate of pressure decrease in the diastolic phase(-dp/dtmax) were decreased, while left ventricular end-diastolic pressure(LVEDP) and level of TNF-α were increased in groups of MI,COA and IS(3. LVW/HW and the increase of TNF-α in group COA were higher than those in groups of MI and ISO(P〈0. 05). Conclusion The model made by coarctation of abdominal aorta has the most obvious manifestation of CHF. Three CHF models mentioned above have their own advantages and disadvantages and selecting an appropriate model should base on the specific experimental requirements and objectives.
