摘要
目的在 SD大鼠气管内滴入猪胰弹性蛋白酶,复制肺气肿模型,观察鼠肺对弹性蛋白酶的反应。方法 18只雄性 SD大鼠,随机分为注酶组 9只,气管内滴入弹性蛋白酶 25U/100g,对照组 9只,气管内注入生理盐水 3ml/kg。饲养 6个月后处死,测量肺脏体积。图像分析观察肺气肿各项改变。结果体重均无明显差异;离体肺体积注酶组和对照组分别为 (10.83± 1.4)ml和 (9.37± 1.1)ml;P< 0.05,有显著差异;注酶组肺泡间隔断裂肺泡腔融合增大,平均肺泡面积和平均内衬间隔增大,而单位面积肺泡数减少。结论利用弹性蛋白酶气管内滴入,复制的模型符合肺气肿改变。
Objective The aim of this study was to evaluate the reaction of the rat- lung to elastase by a single intratracheal instillation of elastase.Method male Sprague- Dawley(SD) rats with initial body weights of~ 100g were used in this study. Animals were randomly divided into controls(CTL,n=9) and emphysemas(EMP, n=9) groups. The EMP rats received a single intratracheal instill of pancreatic porcine elastase (25IU/100g body wt), and CTL were given 0.3ml normal saline. After 6 month , rats were decapitated, their lung volume were determined.Indexes of emphysemas were radiologically observed . Results No significant differences in body weight were noted between EMP and CTL. In vitro lung volumes were larer in EMP than that in CTLanimals (CTL 9.37± 1.1ml, EMP 10.83± 1.4ml; P< 0.05). Mean linear intercept (MLI) and internal surface area (ISA) in EMP were larger than those in CTL, but the number of alveolar counted per unit area was reduced in EMP.Conclusion Our experimental model which induced by single intratracheal instill of elastase conformed to the changes of emphysema.
